Sandwich ELISA - Human IL-2RB ELISA Kit (CD122) (AB275903)
Interpolated concentrations of native IL-2RB in human serum and plasma samples.
The concentrations of IL-2RB were measured in duplicates, interpolated from the IL-2RB standard curves and corrected for sample dilution. Undiluted samples are as follows : serum 100%, plasma (citrate) 100%, plasma (EDTA) 100% and plasma (heparin) 100%. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean IL-2RB concentration was determined to be 274 pg/mL in neat serum, 138 pg/mL in neat plasma (citrate), 155 pg/mL in neat plasma (EDTA) and 225 pg/mL in neat plasma (heparin).

Sandwich ELISA - Human IL-2RB ELISA Kit (CD122) (AB275903)
Interpolated concentrations of native IL-2RB in human PBMC cell culture supernatant samples treated with or without 1.5% PHA-M for 36 hours.
The concentrations of IL-2RB were measured in duplicates, interpolated from the IL-2RB standard curves and corrected for sample dilution. Undiluted samples are as follows : PHA treated PBMC supernatant 100% and untreated PBMC supernatant 100%. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean IL-2RB concentration was determined to be 186 pg/mL in neat PBMC supernatant PHA-M treated and undetectable in untreated PBMC supernatant.

Product details
Human IL-2RB ELISA kit (ab275903) is a single-wash 90 min sandwich ELISA designed for the quantitative measurement of Human IL-2RB protein in human serum, plasma and cell culture supernatant. It uses our proprietary SimpleStep ELISA® technology. Quantitate Human IL-2RB with 8 pg/mL sensitivity.
SimpleStep ELISA® technology employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. See the SimpleStep ELISA® protocol summary in the image section for further details. Our SimpleStep ELISA® technology provides several benefits:
-Single-wash protocol reduces assay time to 90 minutes or less
-High sensitivity, specificity and reproducibility from superior antibodies
-Fully validated in biological samples
-96-wells plate breakable into 12 x 8 wells strips
A 384-well SimpleStep ELISA® microplate (ab203359) is available to use as an alternative to the 96-well microplate provided with SimpleStep ELISA® kits.
IL-2RB (Interleukin-2 receptor subunit beta), also known as CD122, is a receptor for interleukin-2. IL-2 receptor exists in 3 different forms: a high affinity non-covalent dimer of an alpha and a beta subunit, an intermediate affinity monomer of beta subunit, and a low affinity monomer of alpha subunit. The high and intermediate affinity forms can also associate with a gamma subunit. The beta subunit is involved in receptor mediated endocytosis and transduces the mitogenic signals of IL2.

Biological function summary
IL2R beta plays a critical role in cytokine binding and signal transduction for the interleukin-2 receptor which is a heterodimeric complex. This complex consists of the IL-2 receptor alpha (CD25) IL-2 receptor beta and a common gamma chain (γc CD132). The presence of IL-2R beta allows the receptor to mediate biological processes like cell proliferation differentiation and survival particularly within the immune system. The extent to how IL2R beta integrates into these processes makes it an important player in immune regulation and homeostasis.
Pathways
IL2R beta serves essential functions in the JAK-STAT and PI3K-AKT pathways. The binding of IL-2 to its receptor initiates a cascade of intracellular events which involve Janus kinases (JAKs) and signal transducer and activator of transcription (STATs) leading to transcriptional activation of various genes. Along with these pathways IL2R beta also interacts collaboratively with the gamma chain (γc) in promoting Downstream signaling events which enhance cell proliferation and survival.
