Interpolated concentrations of spike IL33 in human serum, plasma and cell culture media (RPMI + 10% FBS) samples.
The concentrations of IL33 were measured in duplicates, interpolated from the IL33 standard curves and corrected for sample dilution. Undiluted samples are as follows : serum neat, plasma (heparin) neat, plasma (EDTA) neat and RPMI + 10% FBS neat. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2).

Interpolated concentrations of native IL33 in human PMA/LPS stimulated THP-1 cell extract based on a 1,000 μg/mL extract load.
The concentrations of IL33 were measured in duplicate and interpolated from the IL33 standard curve and corrected for sample dilution. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean IL33 concentration was determined to be 351.5 pg/mL in PMA/LPS stimulated THP-1 cell extract and undetectable in unstimulated THP-1 cell extract (not shown).

Comparison of IL33 in unstimulated and PMA/LPS stimulated THP-1 cell supernatants.
THP-1 cells were cultured for 24 hours in the presence or absence of 50 ng/mL PMA and 10 μg/mL LPS. The concentrations of IL33 were measured in neat supernatant samples in quadruplicates and interpolated from the IL33 standard curve. The interpolated values are plotted (mean +/- SD, n=3). The mean IL33 concentration was determined to be 15.95 pg/mL in neat PMA/LPS stimulated THP-1 cell culture supernatant and undetectable in neat unstimulated THP-1 cell culture supernatant and RPMI media (not shown).

Human IL-33 ELISA Kit ab223865 is a rapid single-wash 90-min Sandwich ELISA to measure Human IL-33 in cell culture extracts, cell culture supernatant, plasma, serum. This SimpleStep sensitivity is 4.7 pg/mL.
How the assay works
Human IL-33 SimpleStep ELISA®employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. 1. Mix: add samples/standards to the wells together with the capture and detector antibody cocktail. Incubate 1 hr at room temperature
2. Wash
3. Add TMB development solution - incubate for 10 min
4. Add Stop solution
5. Read the results on a plate reader at 450 nm

IL-33 (Interleukin-33, also known as IL-1F11 or NF-HEV) is a proinflammatory cytokine member of the IL-1 family. It is widely expressed and functions by binding and signaling through the IL-1RL1/ST2 receptor. IL-33 is expressed as a pro-protein; the antibodies in this kit were generated using the mature form of IL-33 (aa 109-270).
