Notes

Human IL-6 ELISA kit (ab178013) is a single-wash 90 min sandwich ELISA designed for the quantitative measurement of human IL-6 in serum, plasma and cell culture supernatant.

How the assay works

Mix, Wash, Read : get results in 90 minutes

  1. 1. The sample is added to the plate with the capture and detector antibodies, allowing a sandwich complex to form in solution in a single step.
  2. 2. The capture antibody is conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. After a 60 min incubation the plate is washed.
  3. 3. TMB development solution is added and then stop solution. The plate is then read at 450nm.

Our SimpleStep ELISA® technology provides several benefits:

  • - Single-wash protocol reduces assay time to 90 minutes or less
  • - High sensitivity, specificity and reproducibility from superior antibodies
  • - Fully validated in biological samples
  • - 96-wells plate breakable into 12 x 8 wells strips

Different formats for different needs:

Human IL-6 ELISA kit (ab178013) is available in 3 different formats to adapt to every need:

  • - Single 96 plate kit
  • - 10 x 96 plates bulk size kit: buy the same lot in bulk
  • - 384-well plate kit: increase your throughput with the same easy protocol and save on precious samples.

Looking for more flexibility? The antibody pair used in our kit is available in a carrier-free format, free from BSA and Azide, ready for conjugation:

ab243973: Human/Monkey IL-6 antibody pair kit – BSA and Azide free

ab244758: Anti-IL-6 antibody [IL-6-10] - BSA and Azide free (Capture)

ab244886: Anti-IL-6 antibody [IL-6-5] - BSA and Azide free (Detector)

Performance

ASSAY SPECIFICITY This kit recognizes both native and recombinant Human IL-6 protein in serum, plasma, and cell culture supernatant samples only. Cell and tissue extract samples have not been tested with this kit.

CROSS REACTIVITY Recombinant Human IL-6Ra was prepared at 50 ng/mL and 500 pg/mL and assayed for cross reactivity. No cross-reactivity was observed.

INTERFERENCE Recombinant Human IL-6Ra was prepared at 50 ng/mL and 500 pg/mL and tested for interference. No interference with was observed.

SPECIES REACTIVITY This kit recognizes Human IL-6 protein. Recombinant mouse IL-6 was prepared at 500 pg/mL and assayed for cross reactivity. No cross-reactivity was observed. Please contact our Technical Support team for more information.

About IL-6

Interleukin 6 (IL6) is a cytokine with a wide variety of biological functions. It is a potent inducer of the acute phase of the immune response and plays an essential role in the final differentiation of B-cells into Ig-secreting cells. IL-6 is involved in lymphocyte and monocyte differentiation and IL-6 induces myeloma and plasmacytoma growth as well as nerve cells differentiation. B-cells, T-cells, hepatocytes, hematopoeitic progenitor cells and cells of the CNS are all responsive to IL-6. IL-6 is discharged into the bloodstream after muscle contraction and acts to increase the breakdown of fats and to improve insulin resistance.

Involvement in diseases:

IL-6 is involved in several disease pathways mainly through its pro-inflammation properties.

IL-6 is a mediator of inflammation is several autoimmune conditions including rheumatoid Arthritis (RA), Systemic Lupus Erythematosus (SLE), Multiple Sclerosis (MS). In Type 1 Diabetes: IL-6 plays a role in the autoimmune destruction of pancreatic beta cells.

IL-6 also mediates inflammation in infectious Diseases such as Sepsis or COVID-19.

Insome Cancers (breast, prostate and ovarian cancer), elevated IL-6 levels are associated with poor prognosis. IL-6 also acts as a growth factor for myeloma cells in multiple myeloma promoting their proliferation.

In metabolic disorders like obesity or type 2 diabetes, IL-6 is also linked to chronic inflammation.

In Cardiovascular Diseases: IL-6 promotes Atherosclerosis progression and elevated IL-6 levels are associated with adverse outcomes in heart failure patients.

IL-6 is also involved in Neurodegenerative Diseases such as Alzheimer’s disease by contributing to neuroinflammation. Check our Alzheimer’s disease research tools here.

In cell and gene therapy (CGT) and drug discovery, measuring cytokine release like IL-6 is essential for optimizing therapeutic regimens, monitoring patient responses, and ensuring the overall success of cell and gene therapy interventions.
