Interpolated concentrations of native IL-6 R alpha in human serum, plasma (EDTA), plasma (citrate), plasma (heparin).
The concentrations of IL-6 R alpha were measured in duplicates, interpolated from the target standard curves and corrected for sample dilution. Undiluted samples are as follows : serum 10%, plasma (EDTA) 20%, plasma (citrate, plasma - heparin. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean target concentration was determined to be 34.0 ng/mL in serum, 26.1 ng/mL in plasma (EDTA), 40.8 ng/mL in plasma (citrate), and 33.0 ng/mL in plasma (heparin).

Serum from eight individual healthy female donors was measured in duplicate.
Interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean IL-6 R alpha concentration was determined to be 31.6 ng/mL with a range of 22.4 – 45.1 ng/mL.

Human IL-6 alpha ELISA kit (ab2778117) is a single-wash 90 min sandwich ELISA designed for the quantitative measurement of Human IL-6 alpha protein in human serum, plasma and cell culture supoernatant samples. It uses our proprietary SimpleStep ELISA® technology. Quantitate Human IL-6 alpha with 76.3 pg/mL sensitivity.
SimpleStep ELISA® technology employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. Interleukin-6 Receptor (IL-6R, IL-6 Receptor), also known as Interleukin-6 Receptor subunit alpha and membrane glycoprotein 80, is part of a receptor that binds Interleukin-6 (IL-6). IL-6 Receptor weakly binds IL-6 with a low affinity, and then is presented to gp130 to facilitate transition into a high affinity signaling competent hexamer. Activation may lead to regulation of the immune response, acute-phase reactions and hematopoiesis. The production of IL-6 Receptor has been implicated in the pathogenesis of many diseases, including multiple myeloma, auto immune diseases and prostate cancer.IL-6 Receptor has two isoforms. The first is a transmembrane protein in which the N-terminal extracellular portion can be released by proteolytic cleavage. The second isoform is a secreted form that lacks the transmembrane and intracellular portions. This assay is directed at the extracellular portion of IL-6 Receptor and detects both the soluble and membrane bound forms.

Biological function summary
IL-6 receptor function involves mediating the effects of IL-6 through its interaction with glycoprotein 130 (gp130) forming a signaling complex. This complex initiates intracellular pathways that lead to gene expression changes necessary for immune responses inflammation and hematopoiesis. When IL-6 binds IL-6R the complex formation with gp130 triggers downstream signaling that impacts cell growth differentiation and survival.
Pathways
IL-6R is central to the JAK/STAT signaling pathway one of the two main pathways activated by IL-6 signaling. It's also involved in the MAPK cascade contributing to cellular responses such as proliferation and differentiation. Through these pathways IL-6R interacts with other proteins like JAK kinases and STAT transcription factors coordinating cellular responses to external signals.
