Interpolated concentration of native iNOS was measured in duplicate at different sample concentrations. Undiluted samples are as follows : Caco-2 cell extract 800 µg/mL and Lung tissue extract 750 µg/mL. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). Sample dilutions are made in 1X Cell Extraction Buffer PTR.

Interpolated concentrations of native iNOS in Caco-2 cell extract samples based on an 800 μg/mL extract load.

The concentrations of iNOS were measured in duplicate and interpolated from the iNOS standard curve and corrected for sample dilution. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean iNOS concentration was determined to be 613.7 pg/mL in Caco-2 cell extract samples.

Human iNOS ELISA Kit, Chemiluminescent (ab326909) is a rapid single-wash sandwich 55-min ELISA to measure Human iNOS in Cell Lysate, Tissue Extract with a sensitivity of 1.37 pg/mL.

How the assay works

Human iNOS ELISA Kit, Chemiluminescent (ab326909) SimpleStep Ignite™ ELISA employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step during a 30 min only incubation, significantly reducing assay time. By utilizing a chemiluminscent detection method this improves the sensitivity and dynamic range when compared to traditional colorimetric ELISAs.

Target data

Produces nitric oxide (NO) which is a messenger molecule with diverse functions throughout the body (PubMed : 7504305, PubMed : 7531687, PubMed : 7544004, PubMed : 7682706). In macrophages, NO mediates tumoricidal and bactericidal actions. Also has nitrosylase activity and mediates cysteine S-nitrosylation of cytoplasmic target proteins such PTGS2/COX2 (By similarity). As component of the iNOS-S100A8/9 transnitrosylase complex involved in the selective inflammatory stimulus-dependent S-nitrosylation of GAPDH on 'Cys-247' implicated in regulation of the GAIT complex activity and probably multiple targets including ANXA5, EZR, MSN and VIM (PubMed : 25417112). Involved in inflammation, enhances the synthesis of pro-inflammatory mediators such as IL6 and IL8 (PubMed : 19688109).
