Integrin alpha-2/beta-1 is a receptor for laminin, collagen, collagen C-propeptides, fibronectin and E-cadherin. It recognizes the proline-hydroxylated sequence G-F-P-G-E-R in collagen. It is responsible for adhesion of platelets and other cells to collagens, modulation of collagen and collagenase gene expression, force generation and organization of newly synthesized extracellular matrix. Serum of ten individual healthy human female donors was measured in duplicate. Interpolated dilution factor corrected values (to neat sample) are plotted (mean +/- SD, n=2). The mean Integrin alpha 2 concentration was determined to be 75.12 ng/mL with a range of 34.54 – 103.82 ng/mL.
Interpolated concentration of native Integrin alpha 2 was measured in duplicate at different sample concentrations. Undiluted samples are as follows: serum 1:20, plasma (citrate) 1:25, plasma (EDTA) 1:67, and plasma (heparin) 1:33. The interpolated dilution factor corrected values (to neat sample) are plotted (mean +/- SD, n=2). Sample dilutions are made in Sample Diluent NS.
The concentration of Integrin alpha 2 was measured in wild-type and knockout HeLa cell extracts based on a 100 µg/mL extract load in 1X Cell Extraction Buffer PTR (not provided with this kit, but available as Extraction Buffer 5X PTR ab193970). The mean concentration was determined to be 2,953.1 pg/mL in wild-type cell extract and undetectable in ITGA2 knockout cell extract. The wild-type and Integrin alpha 2 knockout cell extracts are available as Human ITGA2 (CD49b) knockout HeLa cell lysate ab257262.
