Interpolated concentrations of native Interferon beta in human A549 cell culture supernatant samples transfected with 400 ng/mL or 200 ng/mL Poly I : C for 24 hours.
The concentrations of Interferon beta were measured in duplicates, interpolated from the Interferon beta standard curves and corrected for sample dilution. Undiluted samples are as follows : A549 cell culture supernatant transfected with 400 ng/mL Poly I : C 100%, A549 cell culture supernatant transfected with 200 ng/mL Poly I : C 100%. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean Interferon beta concentration was determined to be 111.4 pg/mL in neat A549 supernatant transfected with 400 ng/mL Poly I : C and 33.7 pg/mL in neat A549 supernatant transfected with 200 ng/mL Poly I : C. There was no detectable signal in mock treated A549 supernatant samples.

How the assay works

Human Interferon beta is a 166 amino acids long secreted cytokine that is produced by fibroblasts, dendritic cells, macrophages, and endothelial cells upon pathogen exposure. Interferon beta is type I interferon which mainly helps to stimulate anti-viral response. Interferon beta signals through the heterodimeric IFN-alpha/beta Receptor and activates the JAK/STAT signaling pathway.

Biological function summary
Interferon beta functions as an important mediator of immune functions regulating the activity of natural killer cells and macrophages. It also boosts antigen presentation to T cells. Interferon beta belongs to the larger family of interferons which includes IFN-alpha and IFN-gamma each having distinct effects but working together to mount an effective immune response. Though it does not form part of a complex it plays a critical standalone role in immune signaling pathways.
Pathways
Interferon beta associates strongly with the JAK-STAT signaling pathway. Upon activation it interacts with receptors to phosphorylate STAT proteins mainly STAT1 and STAT2 which then dimerize and translocate to the nucleus to trigger gene expression. Another important pathway is the antiviral response where interferon beta modulates the expression of hundreds of interferon-stimulated genes. These pathways intricately involve other proteins like IFN-alpha and IFN-gamma which work synergistically to establish an effective antiviral environment.
