Sandwich ELISA - Human Kallikrein 10 ELISA Kit (AB279406)
Serum from ten individual healthy human female donors was measured in duplicate.
Interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean Kallikrein 10 concentration was determined to be 1,062.93 pg/mL with a range of 728.05 – 1,332.52 pg/mL.

Sandwich ELISA - Human Kallikrein 10 ELISA Kit (AB279406)
Interpolated concentrations of native Kallikrein 10 in human serum and plasma samples.
The concentrations of Kallikrein 10 were measured in duplicates, interpolated from the Kallikrein 10 standard curves and corrected for sample dilution. Undiluted samples are as follows : serum 100%, plasma (citrate) 50%, plasma (heparin) 100%, and plasma (EDTA) 100%. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean Kallikrein 10 concentration was determined to be 263.09 pg/mL in serum, 526.64 pg/mL in plasma (citrate), 380.88 pg/mL in plasma (heparin), and 195.75 pg/mL in plasma (EDTA).

Product details
Human Kallikrein 10 ELISA kit (ab279406) is a single-wash 90 min sandwich ELISA designed for the quantitative measurement of Human Kallikrein 10 protein in human serum, plasma, and cell culture supernatant samples. It uses our proprietary SimpleStep ELISA® technology. Quantitate Human Kallikrein 10 with 5.29 pg/mL sensitivity.
SimpleStep ELISA® technology employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. See the SimpleStep ELISA® protocol summary in the image section for further details. Our SimpleStep ELISA® technology provides several benefits:
-Single-wash protocol reduces assay time to 90 minutes or less
-High sensitivity, specificity and reproducibility from superior antibodies
-Fully validated in biological samples
-96-wells plate breakable into 12 x 8 wells strips
A 384-well SimpleStep ELISA® microplate (ab203359) is available to use as an alternative to the 96-well microplate provided with SimpleStep ELISA® kits.
Kallikrein 10 is a member of a family of serine proteases that are involved in a variety of cellular functions. Kallikrein 10 has been linked to carcinogenesis as having a tumor suppressor role in breast and prostate cancer. Kallikrein 10 is upregulated in gastrointestinal cancers but downregulated in breast cancer, prostate cancer, NSC lung cancer and renal call carcinoma.

Biological function summary
Kallikrein 10 is part of a larger family of serine proteases involved in various physiological processes. These proteases participate in tissue homeostasis and in some instances immune response. KLK10 expression links to cell proliferation and survival potentially influencing cancer progression. The network of serine proteases forms a proteolytic cascade where multiple proteases sequentially activate amplifying the biological responses.
Pathways
Kallikrein 10 plays a critical role in the proteolytic cascade responsible for regulating homeostatic processes. It participates notably in pathways such as the kallikrein-kinin system and the regulation of tumor microenvironment. KLK10 interacts with other kallikreins like KLK5 and KLK6 mediating their activation and functioning in tissue remodeling and repair processes.
