The Human Kallikrein 11 (KLK11) Enzyme-Linked Immunosorbent Assay (ELISA) kit (ab213801) is designed for the quantitative measurement of Human KLK11 in cell culture supernatants, cell lysates, tissue homogenates, serum and plasma (heparin, EDTA).
The ELISA kit is based on standard sandwich enzyme-linked immune-sorbent assay technology. A monoclonal antibody from mouse specific for KLK11 has been pre-coated onto 96-well plates. Standards and test samples are added to the wells, a biotinylated detection polyclonal antibody from goat specific for KLK11 is added subsequently and then followed by washing with PBS or TBS buffer. Avidin-Biotin-Peroxidase Complex is added and unbound conjugates are washed away with PBS or TBS buffer. HRP substrate TMB is used to visualize HRP enzymatic reaction. TMB is catalyzed by HRP to produce a blue color product that changed into yellow after adding acidic stop solution. The density of yellow is proportional to the Human KLK11 amount of sample captured in plate.
Kallikrein 11 is a protein that in humans is encoded by the KLK11 gene. Kallikreins are a subgroup of serine proteases having diverse physiological functions. Growing evidence suggests that many kallikreins are implicated in carcinogenesis and some have potential as novel cancer and other disease biomarkers. This gene is one of the fifteen kallikrein subfamily members located in a cluster on chromosome 19. Alternate splicing of this gene results in two transcript variants encoding two different isoforms which are differentially expressed.
Kallikrein 11 also known as hK11 is a secreted serine protease enzyme encoded by the KLK11 gene. It has an average molecular mass of approximately 30 kDa. This enzyme is mainly produced in various tissues including prostate spleen and thyroid playing a role in proteolytic processing. Kallikrein 11 has a function in post-translational modifications contributing to the activation and regulation of other proteins through cleavage.
Kallikrein 11 functions as a significant participant in seminal fluid production and sperm motility. It plays roles in maintenance and regulation of these physiological processes. Kallikrein 11 is part of the extensive kallikrein-related peptidase family; hence it shares functional characteristics with other serine proteases in this family. It contributes to the breaking down of extracellular matrix proteins implicating it in tissue remodeling.
Kallikrein 11 integrates into the kallikrein-kinin system and affects components involved in this biochemical network. This system impacts blood pressure regulation and inflammation processes. Kallikrein 11 acts in conjunction with other kallikreins and kinins leading to potential modulation of inflammatory responses and vascular functions. Its actions are related to pathways involving proteins such as prostatic acid phosphatase and prostate-specific antigen (PSA).
Kallikrein 11 has been implicated in prostate cancer and Alzheimer's disease. Increased expression of kallikrein 11 correlates with prostate cancer progression and serves as a potential biomarker for this condition. Additionally research suggests an association between aberrant kallikrein activity and Alzheimer's pathology though this involves more intricate enzymatic pathways with proteins like amyloid precursor protein (APP). Understanding its role in these diseases may offer new therapeutic avenues.
