Human Kallikrein 14 ELISA Kit (ab283870) is a single-wash 90 min sandwich ELISA designed for the quantitative measurement of Kallikrein 14 protein in serum, plasma and cell culture supernatant samples. It uses our proprietary SimpleStep ELISA® technology. Quantitate Human Kallikrein 14 with 9.06 pg/ml sensitivity.
SimpleStep ELISA® technology employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. Kallikrein 14 is a serine protease and member of the kallikrein family of proteases. It has been implicated in carcinogenesis and skin disorders. Kallikrein 14 is activated by KLK5, and its activation leads to a downstream activation of additional kallikreins.
Kallikrein 14 (KLK14) a serine protease is involved in various proteolytic activities. It has a molecular mass of approximately 25 kDa. KLK14 is part of the kallikrein-related peptidase family and is expressed in diverse tissues including skin prostate and brain. This protein is also known by the alternate name hK14. The expression pattern of KLK14 suggests its involvement in local tissue-specific functions.
KLK14 influences processes like skin desquamation and seminal plasma liquefaction. It plays a significant role in the regulation of extracellular matrix components and participates in activating protein precursors. Though it functions as an individual protease KLK14 interacts with other kallikreins in a possible cascade-like manner indicating complex interplay between these proteins.
KLK14 is key in processes involving the kallikrein-kinin system and the activation of certain growth factors. It can cleave and activate other proteins such as renin-angiotensin system components influencing blood pressure regulation. KLK14 operates within this intricate network alongside proteins like KLK1 and other kallikreins that have complementary roles in physiological pathways.
KLK14 has been linked to prostate cancer and skin-related conditions such as atopic dermatitis. Overexpression of KLK14 correlates with prostate cancer progression suggesting its involvement in tumorigenesis. It is also associated with other kallikreins like KLK3 (PSA) in cancer influencing disease development and prognosis. In skin disorders KLK14 may affect processes like inflammation and barrier function revealing its therapeutic target potential.

The Kallikrein 14 standard curve was prepared as described in Section 10. Raw data values are shown in the table. Background-subtracted data values (mean +/- SD) are graphed.
Interpolated concentrations of native Kallikrein 14 in human serum and plasma (citrate) samples.
The concentrations of Kallikrein 14 were measured in duplicates, interpolated from the Kallikrein 14 standard curves and corrected for sample dilution. Undiluted samples are as follows: serum 100% and plasma (citrate) 100% The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean Kallikrein 14 concentration was determined to be 149.55 pg/mL in serum and 206.62 pg/mL in plasma (citrate).
Serum from ten individual healthy human female donors was measured in duplicate.
Interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean Kallikrein 14 concentration was determined to be 376.65 pg/mL with a range of 138.87 – 708.87 pg/mL.
