Human KAP1 ELISA Kit (ab284009) is designed for detection of KAP-1 in human plasma, serum, saliva, cell culture, and cell lysate samples. This assay employs a quantitative sandwich enzyme immunoassay technique that measures human KAP-1 in approximately 4 hours.

A polyclonal antibody specific for human KAP-1 has been pre-coated onto a 96-well microplate with removable strips. KAP-1 in standards and samples is sandwiched by the immobilized antibody and a biotinylated polyclonal antibody specific for human KAP-1, which is recognized by a streptavidin-peroxidase (SP) conjugate.The color development is stopped and the intensity of the color is measured.

KAP1 also known as TRIM28 or TIF1β is a multifunctional protein involved in transcriptional regulation. It has a molecular weight of approximately 87 kDa. This protein is expressed in a wide variety of human tissues where it plays a role as a co-repressor for KRAB domain-containing zinc finger proteins. KAP1 recruits various epigenetic modifiers to silence transcription and this activity depends on its phosphorylation status. Biological function summary

KAP1 acts as a scaffold protein facilitating the assembly of large protein complexes that include chromatin remodelers and histone deacetylases. It is significant in maintaining chromatin structure and mediating gene silencing. KAP1 phosphorylation serves as a switch between its roles in transcriptional repression and activation. This protein also interacts with heterochromatin protein 1 (HP1) and other TRIM family proteins playing an important role in genomic stability by controlling gene expression and repair mechanisms.

In neurological disorders dysregulation of KAP1 is associated with impaired neural development and neurodegeneration. NADPH-dependent oxidoreductase which catalyzes the reduction of dicarbonyl compounds. Displays reductase activity in vitro with 3,4-hexanedione, 2,3-heptanedione and 1-phenyl-1,2-propanedione as substrates (PubMed : 16685466). May function as a dicarbonyl reductase in the enzymatic inactivation of reactive carbonyls involved in covalent modification of cellular components (PubMed : 16685466). Also displays a minor hydroxysteroid dehydrogenase activity toward bile acids such as ursodeoxycholic acid (UDCA) and isoursodeoxycholic acid (isoUDCA), which makes it unlikely to control hormone levels (PubMed : 16685466). Doesn't show any activity in vitro with retinoids and sugars as substrates (PubMed : 16685466). Attenuates MDM2-mediated p53/TP53 degradation, leading to p53/TP53 stabilization and increased transcription activity, resulting in the accumulation of MDM2 and CDKN1A/p21 (PubMed : 20547751). Reduces proliferation, migration and invasion of cancer cells and well as the production of ROS in cancer (PubMed : 29106393).
