Kininogens are inhibitors of thiol proteases. HMW-kininogen plays an important role in blood coagulation by helping to position optimally prekallikrein and factor XI next to factor XII; HMW-kininogen inhibits the thrombin- and plasmin-induced aggregation of thrombocytes. LMW-kininogen inhibits the aggregation of thrombocytes. LMW-kininogen is in contrast to HMW-kininogen not involved in blood clotting. Bradykinin. The active peptide bradykinin is a potent vasodilatator that is released from HMW-kininogen shows a variety of physiological effects: (A) influence in smooth muscle contraction, (B) induction of hypotension, (C) natriuresis and diuresis, (D) decrease in blood glucose level, (E) it is a mediator of inflammation and causes (E1) increase in vascular permeability, (E2) stimulation of nociceptors (4E3) release of other mediators of inflammation (e.g. prostaglandins), (F) it has a cardioprotective effect (directly via bradykinin action, indirectly via endothelium-derived relaxing factor action).

Abcam's Kininogen (KNG) Human in vitro ELISA (Enzyme-Linked Immunosorbent Assay) kit is designed for the quantitative measurement of Kininogen in urine, saliva, milk, cerebrospinal fluid and cell culture supernatants.
A Kininogen specific antibody has been precoated onto 96-well plates and blocked. Standards or test samples are added to the wells and subsequently a Kininogen specific biotinylated detection antibody is added and then followed by washing with wash buffer. Streptavidin-Peroxidase Conjugate is added and unbound conjugates are washed away with wash buffer. TMB is then used to visualize Streptavidin-Peroxidase enzymatic reaction. TMB is catalyzed by Streptavidin-Peroxidase to produce a blue color product that changes into yellow after adding acidic stop solution. The density of yellow coloration is directly proportional to the amount of Kininogen captured in plate.

The kininogen is a protein involved in various biological processes and also known as high molecular weight kininogen (HMWK). This protein which has a mass of approximately 120 kDa circulates in the plasma. Kininogen is synthesized by the liver and endothelial cells where it exists as HMWK. It serves as a precursor to kinins like bradykinin which are important in regulating blood pressure and inflammation.
Kininogen participates in the kallikrein-kinin system. It plays an important role in blood coagulation and the inflammatory response. HMWK is not part of a complex but interacts with factors such as prekallikrein and Factor XII. By serving as a co-factor HMWK facilitates the activation of these molecules which progresses the coagulation cascade and inflammatory pathways.
The kininogen functions within the kallikrein-kinin and coagulation systems. It interacts closely with enzymes like kallikrein which cleaves kininogen to release bradykinin assisting in vasodilation and increasing vascular permeability. Additionally HMWK is tethered to the intrinsic pathway of coagulation by activation of Factor XII which links it to proteins involved in blood clot formation.
The kininogen is associated with hereditary angioedema and bradykinin-mediated edema. Abnormalities in its function can lead to excessive bradykinin release. This condition involves excessive swelling due to alterations in vascular permeability. Moreover the protein is linked to chronic inflammatory diseases through its connection with the bradykinin receptor and related peptides.
