Interpolated concentrations of native KLF4 in human cell extract based on a 500 μg/mL extract load.

The concentrations of KLF4 were measured in duplicate and interpolated from the KLF4 standard curve and corrected for sample dilution. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean KLF4 concentration was determined to be 417.5 pg/mL in NCCIT extract.

How the assay works

Human KLF4 SimpleStep ELISA^{^®}employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA^{^®} plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. KLF4, also known as Krueppel-like factor 4, is an embryonic transcription factor involved in the maintenance of maintaining pluripotency by preventing differentiation. It is also found in post-mitotic cells, where its overexpression inhibits proliferation. KLF4 is currently under investigation as a tumor suppressor given it potent ability to halt proliferation. Altered expression of its gene has been noted in many types of human cancers.
