Interpolated concentrations of native KLF4 in human cell extract based on a 500 μg/mL extract load.
The concentrations of KLF4 were measured in duplicate and interpolated from the KLF4 standard curve and corrected for sample dilution. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean KLF4 concentration was determined to be 417.5 pg/mL in NCCIT extract.

How the assay works
Human KLF4 SimpleStep ELISA®employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. 1. Mix: add samples/standards to the wells together with the capture and detector antibody cocktail. Incubate 1 hr at room temperature
2. Wash
3. Add TMB development solution - incubate for 10 min
4. Add Stop solution
5. Read the results on a plate reader at 450 nm

KLF4, also known as Krueppel-like factor 4, is an embryonic transcription factor involved in the maintenance of maintaining pluripotency by preventing differentiation. It is also found in post-mitotic cells, where its overexpression inhibits proliferation. KLF4 is currently under investigation as a tumor suppressor given it potent ability to halt proliferation. Altered expression of its gene has been noted in many types of human cancers.

Biological function summary
KLF4 impacts cell differentiation and proliferation through its role as a transcription factor. It functions within several cellular complexes and interacts with other transcription factors to modulate gene expression. KLF4 regulates stem cell maintenance by influencing gene networks involved in self-renewal and differentiation contributing particularly to pluripotency in embryonic stem cells.
Pathways
The transcriptional activity of KLF4 integrates into key signaling pathways like the Wnt/?-catenin and Notch pathways. Within the Wnt/?-catenin pathway KLF4 may cooperate with ?-catenin and TCF/LEF transcription factors to modulate gene targets. Similarly in the Notch signaling pathway KLF4 interacts with pathway components to influence cell fate decisions. These interactions highlight KLF4's central role in orchestrating complex signaling networks.
