Interpolated concentrations of native LAMC2 in human cell culture supernatant samples.

The concentrations of LAMC2 were measured in duplicates, interpolated from the LAMC2 standard curves and corrected for sample dilution. Undiluted samples are as follows : A431 supernatant 1 : 100 and A549 supernatant 5%. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean LAMC2 concentration was determined to be 86,513 pg/mL in A431 supernatant, 3,106 pg/mL in A549 supernatant.

Interpolated concentrations of native LAMC2 in human serum, plasma samples.

The concentrations of LAMC2 were measured in duplicates, interpolated from the LAMC2 standard curves and corrected for sample dilution. Undiluted samples are as follows : serum 50%, plasma (citrate) 25%, plasma (EDTA) neat, and plasma (heparin) 50%. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean LAMC2 concentration was determined to be 368.1 pg/mL in serum, 366.7 pg/mL in plasma (citrate) and 274.7 pg/mL in plasma (EDTA), and 463.9 pg/mL plasma (heparin).

Human LAMC2 ELISA kit (ab282301) is a single-wash 90 min sandwich ELISA designed for the quantitative measurement of Human LAMC2 protein in human serum, plasma, cell culture supernatant, and cell extract samples. It uses our proprietary SimpleStep ELISA® technology. Quantitate Human LAMC2 with 2.16 pg/mL sensitivity.

SimpleStep ELISA® technology employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. See the SimpleStep ELISA® protocol summary in the image section for further details. Our SimpleStep ELISA® technology provides several benefits:

-Single-wash protocol reduces assay time to 90 minutes or less

LAMC2 is the gamma subunit of Laminin 5. Laminin's are complex glycoproteins consisting of an alpha, beta and gamma chain. LAMC2 secreted in lung cancer and associated with the progression of the disease. ab282301 recognizes a fragment of the gamma-2 subunit.

The LAMC2 protein also known as laminin subunit gamma-2 is an essential component of the extracellular matrix. It is a subunit of the laminin-332 complex formerly referred to as laminin-5 and has an approximate mass of 160 kDa. LAMC2 is widely expressed in various tissues most prominently in epithelial tissues. It localizes at the basement membrane zone where epithelial and mesenchymal tissues meet.
Biological function summary

LAMC2 plays a critical role in cell adhesion migration and signal transduction. Laminin-332 which includes LAMC2 binds to integrin receptors on cell surfaces facilitating cell-matrix interactions and communication. This complex is important for basement membrane stability and tissue organization and it is actively involved in wound healing by promoting keratinocyte migration and melanin transport.

Pathways

LAMC2 integrates into several important extracellular matrix pathways including the integrin signaling pathway. These pathways involve interactions with proteins like integrin alpha-6/beta-4 which are linked to cellular adhesion and mobility. LAMC2 also contributes to epithelial cell functions and is involved in the PI3K/AKT signaling pathway highlighting its importance in cell proliferation and survival.

LAMC2 has significant associations with conditions like dystrophic epidermolysis bullosa and invasive carcinomas. In dystrophic epidermolysis bullosa mutations in LAMC2 disrupt the integrity of the basement membrane leading to fragile skin and blistering. In cancer abnormal expression or alteration of LAMC2 can promote tumor invasion and metastasis often in conjunction with overexpression of matrix metalloproteinases (MMPs) that degrade the extracellular matrix therefore facilitating cancer cell infiltration.
