Sandwich ELISA - Human LBP ELISA Kit (LPS Binding Protein) (AB279407)
Serum from ten individual healthy human female donors was measured in duplicate.
Interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean LBP concentration was determined to be 3.11 μg/mL with a range of 0.94 – 4.60 μg/ml.

Sandwich ELISA - Human LBP ELISA Kit (LPS Binding Protein) (AB279407)
Interpolated concentrations of native LBP in human serum and plasma samples.
The concentrations of LBP were measured in duplicates, interpolated from the LBP standard curves and corrected for sample dilution. Undiluted samples are as follows : serum 1 : 4,000, plasma (citrate) 1 : 2,000, plasma (heparin) 1 : 2,000, and plasma (EDTA) 1 : 2,000. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean LBP concentration was determined to be 2.00 μg/mL in serum, 1.39 μg/mL in plasma (citrate), 1.01 μg/mL in plasma (heparin), and 1.50 μg/ml in plasma (EDTA).

Product details
Human LBP ELISA Kit (LPS Binding Protein) ab279407 is a rapid single-wash 90-min Sandwich ELISA to measure Human LBP (LPS Binding Protein)in cell culture media, citrate plasma, EDTA plasma, heparin plasma, serum. This SimpleStep sensitivity is 6.79 pg/mL.
How the assay works
Human LBP (LPS Binding Protein)SimpleStep ELISA®employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. See the SimpleStep ELISA® protocol summary in the image section for further details.
Assay Specificity
Our SimpleStep ELISA® kits use recombinant monoclonal antibodies rigorously validated to ensure the highest level of consistency and reproducibility, improved sensitivity and specificity and ease of scalability and security of supply.
Please refer to our protocol booklet for more details.

1. Mix: add samples/standards to the wells together with the capture and detector antibody cocktail. Incubate 1 hr at room temperature
2. Wash
3. Add TMB development solution - incubate for 10 min
4. Add Stop solution
5. Read the results on a plate reader at 450 nm
LBP (LPS Binding Protein) is an acute phase protein involved in the cellular response to gram-negative bacterial infections. LBP binds to LPS protein expressed on gram-negative bacteria and elicits a response by engaging with pattern recognition receptors, CD14 and TLR4. LBP is essential for the initial response to gram-negative bacterial infection but not for the downstream stages.

Biological function summary
LBP serves a significant role in enhancing the sensitivity of the immune system to bacterial endotoxins. It does not form a permanent part of a larger complex but works closely with other molecules like CD14 and MD-2 an accessory protein of the TLR4 receptor to mediate the innate immune response. By efficiently binding LPS LBP aids in transporting these molecules to cell surface receptors accelerating the detection and subsequent response to infections.
Pathways
LBP is heavily involved in the TLR4 signaling pathway which is important for the detection and response to Gram-negative bacterial infections. Through this pathway LBP and associated proteins such as CD14 and MD-2 help activate TLR4 triggering downstream NF-κB signaling. This activation leads to the production of pro-inflammatory cytokines and an upsurge in antimicrobial activities fortifying the body's defenses against bacterial invasion.
