Comparison of LC3A in mock-treated and chloroquine-treated HeLa cell extracts. HeLa cells were cultured for 24 hours in the absence (mock) or presence of 50 mM chloroquine. The concentrations of LC3A were measured in three different dilutions of the supernatant samples in duplicates and interpolated from the LC3A standard curve. The interpolated values are plotted (mean +/- SD, n=3). The mean LC3A concentration was determined to be 3.16 ng/mg in mock-treated HeLa cell extract, and 5.64 ng/mg in chloroquine-treated HeLa cell extract.

Interpolated concentrations of native LC3A in human SHSY-5Y cell extract based on a 30 μg/mL extract load, and HeLa cell extract based on a 500 μg/mL extract load. The concentrations of LC3A were measured in duplicate and interpolated from the LC3A standard curve and corrected for sample dilution. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean LC3A concentration was determined to be 2,048 pg/mL in SHSY-5Y cell extract and 1,601 pg/mL in HeLa cell extract.

Human LC3A ELISA Kit (ab239432) is a single-wash 90 min sandwich ELISA designed for the quantitative measurement of LC3A protein in cell culture extracts. It uses our proprietary SimpleStep ELISA® technology. Quantitate Human LC3A with 2.1 pg/ml sensitivity.

SimpleStep ELISA® technology employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. See the SimpleStep ELISA® protocol summary in the image section for further details. Our SimpleStep ELISA® technology provides several benefits:

The protein LC3A also known as Microtubule-associated proteins 1A/1B light chain 3A plays an important role in the autophagy process. It exists in two forms LC3-I and LC3-II with the latter being lipidated and integrated into autophagosomal membranes. LC3A has a molecular mass of approximately 17 kDa. Researchers observe its expression in various tissues but most highly in muscle liver and brain. It is often studied along with LC3B due to their similar roles in autophagy.
Biological function summary

LC3A participates in the formation of autophagosomes which are double-membraned vesicles that capture cellular waste for degradation. LC3A's conversion from LC3-I to LC3-II on the autophagosomal membrane signals the initiation of autophagy. It associates with proteins in the Atg family forming complexes that regulate the maturation of autophagosomes. The LC3 family including LC3A and LC3B acts as important components in the cell's ability to maintain homeostasis through autophagic activity.

Pathways

LC3A plays a vital part in the autophagy pathway and is also linked to the mTOR signaling pathway. Autophagy is a process that helps with the recycling of cellular components and mTOR signaling inhibits autophagy when nutrients are abundant. LC3A's interactions with other proteins such as p62 and ULK1 are essential in modulating these processes and ensuring cell survival under stress conditions. LC3A and LC3B's roles are integral in coordinating the cellular response to nutrient status and stress highlighting their importance in cellular regulation.

LC3A has significant connections to cancer and neurodegenerative diseases. Altered autophagic activity where LC3A takes part appears in various forms of cancer indicating its role in tumor progression and apoptosis regulation. Additionally abnormal LC3A function has implications in disorders such as Alzheimer's disease where autophagic dysfunction leads to neurodegeneration. In these contexts LC3A's relationship with proteins like p62 which accumulate in neurodegenerative diseases provides insight into pathological mechanisms and potential therapeutic targets.
