Human LIF ELISA Kit is a single-wash 90-min Simplestep used to quantify Human LIF with a sensitivity of 1.469 pg/ml. The assay uses a simple mix-wash-read protocol with just one incubation and one wash step.
- Colorimetric Sandwich ELISA - 450 nm readout : works on any standard plate reader
- Different formats for different needs: 384-well plate for higher throughput
- Design your own immunoassay: we also offer the conjugation-ready antibody pair

Sandwich ELISA - Human LIF ELISA Kit (AB242228)
Interpolated concentrations of native LIF in human cell culture supernatant samples. The concentrations of LIF were measured in duplicates, interpolated from the LIF standard curves and corrected for sample dilution. Undiluted samples are 1.5% PHA-M treated PBMC cell culture supernatant 10%. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean LIF concentration was determined to be 8336.44 pg/mL in 1.5% PHA-M treated PBMC cell culture supernatant.

Sandwich ELISA - Human LIF ELISA Kit (AB242228)
Example of human LIF standard curve in Sample Diluent NS. The LIF standard curve was prepared as described in Section 10. Raw data values are shown in the table. Background-subtracted data values (mean +/- SD) are graphed.

Sandwich ELISA - Human LIF ELISA Kit (AB242228)
Interpolated concentration of native LIF was measured in duplicate at different sample concentrations in 96-well vs. 384-well plates. Undiluted samples are 1 : 16 stimulated human PBMC supernatant (1.5% PHA-M for 46 hrs). The interpolated dilution factor corrected values (to neat sample) are plotted (mean +/- SD, n=2). Sample dilutions are made in Sample Diluent NS.

How the assay works
Human LIF SimpleStep ELISA®employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. See the SimpleStep ELISA® protocol summary in the image section for further details.
Assay Specificity
Our SimpleStep ELISA® kits use recombinant monoclonal antibodies rigorously validated to ensure the highest level of consistency and reproducibility, improved sensitivity and specificity and ease of scalability and security of supply.
Please refer to our protocol booklet for more details.

Biological function summary
LIF influences cell differentiation growth and inflammatory response. As part of the interleukin-6 cytokine family LIF exerts its effects through a receptor complex involving LIFR (LIF receptor) and GP130. The LIF protein's signaling controls processes like neuronal cell differentiation and bone formation. It impacts hematopoietic cells and supports embryonic implantation and placental development by preparing the uterine environment.
Pathways
The LIF protein is essential in the JAK/STAT signaling pathway which regulates gene expression vital for survival and proliferation. It also interacts with the MAPK/ERK pathway ensuring diverse cellular responses. These pathways link LIF to related proteins like STAT3 and ERK1/2 which mediate further downstream effects. The interplay between these signaling cascades highlights LIF's role in managing cell survival and proliferation in various contexts.
