Serum from ten individual healthy human male donors was measured in duplicate.
Interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean LRP1 concentration was determined to be 112.6 ng/mL with a range of 66.7 – 236.6 ng/mL.

Interpolated concentrations of native LRP1 in mouse serum and rat serum samples.
The concentrations of LRP1 were measured in duplicates, interpolated from the LRP1 standard curves and corrected for sample dilution. Undiluted samples are as follows : mouse serum 2.5% and rat serum 2.5%. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean LRP1 concentration was determined to be 199.4 ng/mL in mouse serum and 104.9 ng/mL in rat serum.

The concentrations of LRP1 were measured in duplicates, interpolated from the LRP1 standard curves and corrected for sample dilution. Undiluted samples are as follows : serum 5%, plasma (citrate) 5%, and RAW264.7 cell culture supernatant. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean LRP1 concentration was determined to be 90.0 ng/mL in serum, 72.0 ng/mL in plasma (citrate) and 29.7 ng/mL in RAW264.7 cell culture supernatant.

Interpolated concentrations of native LRP1 in human HepG2 and Kidney extract samples and samples based on a 200 µg/mL and 400 μg/mL extract load (respectively).
The concentrations of LRP1 were measured in duplicate and interpolated from the LRP1 standard curve and corrected for sample dilution. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean LRP1 concentration was determined to be 3,095 pg/mL in HepG2 extract and 2,662 pg/mL in Kidney extract.

How the assay works
Human LRP1 SimpleStep ELISA®employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. 1. Mix: add samples/standards to the wells together with the capture and detector antibody cocktail. Incubate 1 hr at room temperature
2. Wash
3. Add TMB development solution - incubate for 10 min
4. Add Stop solution
5. Read the results on a plate reader at 450 nm

Prolow-density lipoprotein receptor-related protein 1 (LRP1) is an endocytic receptor involved in endocytosis and in phagocytosis of apoptotic cells. LRP1 is widely expressed on many cell types including neurons, adipocytes, hepatocytes, vascular smooth muscle cells, fibroblasts, keratinocytes, macrophages, and megakaryocytes. Human LRP1 is an N-glycosylated and sialylated molecule that is cleaved in the Golgi apparatus to produce an 85 kDa transmembrane beta chain and a 515 kDa alpha chain that associates noncovalently with the beta chain but does not itself cross the membrane. The provided protein standard for ab278128 is a fragment of the extracellular domain.
