Supplementary information

Netrin G2 Ligand also known as NGL2 is a synaptic adhesion molecule that weighs approximately 70 kDa. It is predominantly expressed in the central nervous system where it plays an important role in neural development and synaptic function. NGL2 belongs to the family of netrin-G ligands which are known for their involvement in neural network organization. The protein serves as a ligand for netrin-G2 facilitating cell-cell interactions necessary for proper synaptic formation.
Biological function summary

NGL2 contributes significantly to intercellular connectivity by forming part of the synaptic scaffold important for neural communication. It interacts with postsynaptic density proteins leading to stabilization and maturity of synaptic connections. NGL2 functions as a critical component in maintaining synaptic strength and plasticity therefore influencing learning and memory. It operates in conjunction with other synaptic proteins to ensure optimal neurotransmission and signal integration within the brain.

Pathways

NGL2 is intimately involved in the glutamatergic neurotransmission pathway important for synaptic excitability and plasticity. It works together with proteins like SAP102 and PSD-95 which are pivotal in synaptic structure and function. NGL2 also plays a role in the axonal guidance signaling pathway essential for directing neuronal growth and alignment. These pathways integrate the function of NGL2 with broader cellular processes highlighting its significance in neural circuitry.

NGL2 has connections to conditions such as autism spectrum disorders (ASD) and schizophrenia. Dysregulation of NGL2 can lead to aberrant synaptic connections and disruptions in neural communication contributing to the pathogenesis of these disorders. The protein's interaction with neuroligins which are associated with ASD further supports its potential role in disease mechanisms. Understanding NGL2 and its involvement in these diseases opens avenues for therapeutic research targeting synaptic dysfunction.

Product protocols

Target data

Synaptic adhesion protein. Regulates the formation of exitatory synapses through the recruitment of pre-and-postsynaptic proteins. Organize the lamina/pathway-specific differentiation of dendrites. Plays an important role for auditory synaptic responses. Involved in the suppression of glioma (By similarity).
