Sandwich ELISA - Human LYST ELISA Kit (AB275905)
Interpolated concentrations of native LYST in human THP-1 cell extract based on a 300 μg/mL extract load.
The concentrations of LYST were measured in duplicate and interpolated from the LYST standard curve and corrected for sample dilution. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean LYST concentration was determined to be 3,115.82 pg/mL.

Human LYST ELISA kit (ab275905) is a single-wash 90 min sandwich ELISA designed for the quantitative measurement of Human LYST protein in human cell extract samples. It uses our proprietary SimpleStep ELISA® technology. Quantitate Human LYST with 20 pg/mL sensitivity.
SimpleStep ELISA® technology employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. See the SimpleStep ELISA® protocol summary in the image section for further details. Our SimpleStep ELISA® technology provides several benefits:
-Single-wash protocol reduces assay time to 90 minutes or less

LYST, lysosomal trafficking regulator, is a vesicular transport protein. Defects in this protein lead to Chédiak–Higashi syndrome, a lysosomal storage disease that manifests as hypopigmentation, neutropenia, defective white blood cell function, and coagulation defects. The immunogen from which this kit was made represents the BEACH domain of the protein. Although this domain is conserved across many mammalian species, no cross-reactivity was seen.

Biological function summary
LYST directly influences the cellular degradation and recycling process. The protein does not act alone but functions as part of a larger cellular machinery that maintains lysosomal homeostasis. Its role is particularly important in immune cell function where it ensures proper lysosomal delivery and secretion especially in cytotoxic T cells and natural killer cells. These activities are important for granulocyte and lymphocyte regulation ensuring a proper response to infections and cellular stress.
Pathways
The protein LYST is integral in pathways related to vesicular transport such as the endocytic and autophagic pathways. It has a well-documented interaction with proteins such as Rab7 which is important for endosome-to-lysosome trafficking. Additionally LYST interacts with components of the cytoskeleton which further influences lysosome positioning within the cell therefore supporting its central role in cellular logistic pathways.
