Interpolated concentrations of native M-CSF in human saliva urine and A549 cell supernatant samples. The concentrations of M-CSF were measured in duplicates interpolated from the M-CSF standard curves and corrected for sample dilution. Undiluted samples are as follows : saliva 25% urine 1 : 200 and A549 cell supernatant 12.5%. The interpolated dilution factor corrected values are plotted (mean +/- SD n=2). The mean M-CSF concentration was determined to be 0.9 ng/mL in saliva 28.1 ng/mL in urine and 0.89 ng/mL in A549 cell supernatant.

Interpolated dilution factor corrected values are plotted (mean +/- SD n=2). The mean M-CSF concentration was determined to be 27.8 ng/mL with a range of 17.7 - 47.2 ng/mL.

The concentrations of M-CSF were measured in duplicates interpolated from the M-CSF standard curves and corrected for sample dilution. Undiluted samples are as follows : serum 2% plasma (heparin) 2.5% and PBMC supernatant 7.5%. The interpolated dilution factor corrected values are plotted (mean +/- SD n=2). The mean M-CSF concentration was determined to be 11.5 ng/mL in serum 6.7 ng/mL in plasma (heparin) and 2.4 ng/mL in stimulated PBMC supernatant.

How the assay works
Human M-CSF ELISA Kit, Chemiluminescent (ab327294) SimpleStep Ignite™ ELISA employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step during a 30 min only incubation, significantly reducing assay time. By utilizing a chemiluminscent detection method this improves the sensitivity and dynamic range when compared to traditional colorimetric ELISAs.

1. Mix : add samples / standards to the wells together with the capture and detector antibody cocktail. Incubate 30 min at room temperature
2. Wash
3. Add ChemiHRP development solution - incubate for 2 min
4. Read the results on a plate reader capable of luminescence detection
