Interpolated concentrations of native MASPIN in human PC-3, A431, HL-60, and Hela cell extract based on a 10 μg/mL extract load.

The concentrations of MASPIN were measured in duplicate, interpolated from the MASPIN standard curve. The interpolated, dilution factor-corrected values are plotted (mean +/- SD, n=2). The mean MASPIN concentration was determined to be 55 ng/mL, 93 ng/mL, and non-detectable in PC-3, A431, HL-60, and HeLa cell extract, respectively.

The interpolated, dilution factor-corrected values are plotted (mean +/- SD, n=2). The mean MASPIN concentration was determined to be 57.5 ng/mL in PC-3 cell extract.

Human MASPIN ELISA Kit (ab218269) is a single-wash 90 min sandwich ELISA designed for the quantitative measurement of MASPIN protein in cell culture extracts. It uses our proprietary SimpleStep ELISA® technology. Quantitate Human MASPIN with 8 pg/ml sensitivity.

SimpleStep ELISA® technology employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. See the SimpleStep ELISA® protocol summary in the image section for further details. Our SimpleStep ELISA® technology provides several benefits:

MASPIN (Mammary Serine Protease Inhibitor) is a 375 amino acid glycosylated protein encoded by the gene SERPINB5. It belongs to a family of proteins known as serpins (serine protease inhibitor), which function as regulators of protein breakdown by acting as suicide inhibitors of the catalytic activity of proteinases. Inhibitory serpins undergo a transition between a stressed and relaxed confirmation, where the stressed confirmation binds to the catalytic serine residue of the protease. MASPIN is thought to interact with extracellular matrix components, glutathione S-transferase, and interferon regulatory factor 6. It was originally thought to play a role in tumor metastasis, but the exact function is unknown.

MASPIN also known as SERPINB5 is a non-inhibitory serine protease inhibitor with a mass of approximately 42 kDa. It functions mechanically to regulate cell adhesion migration apoptosis and angiogenesis. MASPIN localizes mainly in the cytoplasm and nucleus and expresses predominantly in epithelial cells including those in the mammary gland prostate and keratinocytes in the skin. It acts as a tumor suppressor and researchers often use immunohistochemistry (IHC) to study its expression patterns in various tissues.
Biological function summary

MASPIN influences apoptosis and cell motility playing a critical role in tumor suppression. It does not form part of a complex but interacts directly with extracellular matrix components. MASPIN can inhibit angiogenesis by reducing the proliferation and migration of endothelial cells.
