Standard curve comparison between the original Human MCP3 SimpleStep ELISA (Human MCP3 ELISA Kit (CCL7) ab193769) and current Human MCP3 SimpleStep ELISA (ab322362).
2,800 pg/mL of recombinant human MCP1 (CCL2), MCP2 (CCL8), and Eotaxin (CCL11) were individually tested for interference with 1,400 pg/mL of recombinant human MCP3 (CCL7). Percent reactivity was determined by interpolating the protein concentration from the human MCP3 (CCL7) standard curve and expressing the interpolated concentration as a percentage of the protein concentration tested. No significant interference was observed.
50 ng/mL of recombinant human MCP1 (CCL2), MCP2 (CCL8), and Eotaxin (CCL11) were individually tested for cross reactivity. Percent reactivity was determined by interpolating the protein concentration from the human MCP3 (CCL7) standard curve and expressing the interpolated concentration as a percentage of the protein concentration tested. No cross reactivity was observed.
Interpolated concentration of native MCP3 was measured in duplicate at different sample concentrations in 96-well vs. 384-well plates. Undiluted samples are 20% stimulated PBMC supernatant (1.5% PHA-M stimulation for 46 hrs). The interpolated dilution factor corrected values (to neat sample) are plotted (mean +/- SD, n=2). Sample dilutions are made in Sample Diluent NS.

Interpolated concentration of native MCP3 was measured in duplicate at different sample concentrations. Undiluted samples are as follows: 50% unstimulated PBMC supernatant and 20% stimulated PBMC supernatant (1.5% PHA-M stimulation for 46 hrs). The interpolated dilution factor corrected values (to neat sample) are plotted (mean +/- SD, n=2). Sample dilutions are made in Sample Diluent NS. No signal was detected in unstimulated PBMC supernatant.
