Human MIP5 ELISA Kit (ab217774) is a single-wash 90 min sandwich ELISA designed for the quantitative measurement of MIP5 protein in cell culture extracts, cell culture supernatant, cit plasma, edta plasma, hep plasma, other biological fluids, serum, and tissue extracts. It uses our proprietary SimpleStep ELISA® technology. Quantitate Human MIP5 with 27.1 pg/ml sensitivity.
SimpleStep ELISA® technology employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. MIP5 is a secreted monomeric chemotactic factor that attracts T-cells and monocytes, but not neutrophils, eosinophils, or B-cells. MIP5 acts mainly via CC chemokine receptor CCR1. It also binds to CCR3. MIP5 can be cleaved into 3 chains, MIP5(22-92), MIP5(25-92) and MIP5(29-92). These cleavage products are more potent chemo-attractants than MIP5. They are also found in high levels in synovial fluids from rheumatoid patients.

Macrophage inflammatory protein 5 (MIP-5) also known as CCL15 is a chemokine involved in immune system function. It plays an important role in recruiting and activating certain immune cells such as monocytes and lymphocytes. Its molecular mass is approximately 13.5 kDa. MIP-5 is produced mainly by monocytes and macrophages and can be found in the liver and several other tissues. This protein mediates inflammation by guiding immune cells to sites of infection or injury where they can perform immune defense activities.
MIP-5 is involved in the chemotactic movement of immune cells a process where cells move in response to chemical stimuli. It is not part of a protein complex but interacts dynamically with cell surface receptors to induce cellular responses. MIP-5 triggers cells to release further signaling molecules increasing the inflammatory response's effectiveness. This chemokine creates a highly coordinated immune response by initiating a signaling cascade that helps maintain immune surveillance and response to pathogens.
MIP-5 plays a role in the chemokine signaling pathway where it interacts with specific receptors that lead to cellular migration and activation. It also fits into the cytokine-cytokine receptor interaction pathway influencing a range of cellular responses tied to immune function. MIP-5 shares functional properties with other chemokines like MCP-1 (monocyte chemoattractant protein-1) which similarly attract immune cells and mediate immune responses.
MIP-5 is linked to inflammatory diseases and certain types of cancer. Elevated levels of MIP-5 have been observed in rheumatoid arthritis where it contributes to joint inflammation. It is also associated with hepatocellular carcinoma where it may influence tumor microenvironment and metastasis. In these conditions MIP-5 might interact with other inflammatory molecules such as IL-6 and TNF-alpha suggesting its role as a mediator of chronic inflammation and malignancy processes.

Interpolated concentrations of native MIP5 in human serum and plasma samples.
The concentrations of MIP5 were measured in duplicates, interpolated from the MIP5 standard curves and corrected for sample dilution. Undiluted samples are as follows: serum 20%, plasma (citrate) 20%, plasma (heparin) 20% and plasma (EDTA) 20%. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean MIP5 concentration was determined to be 8705 pg/mL in neat serum, 7196 pg/mL in neat plasma (citrate), 6848 pg/mL in neat plasma (heparin), and 7231 pg/mL in neat plasma (EDTA).
Interpolated concentrations of native MIP5 in HepG2 (4 days) cell culture supernatant and human synovial fluid samples.
The concentrations of MIP5 were measured in duplicates, interpolated from the MIP5 standard curves and corrected for sample dilution. Undiluted samples are as follows: HepG2 cell culture supernatant 100% (neat), and synovial fluid 20%. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean MIP5 concentration was determined to be 706.2 pg/mL in neat HepG2 cell culture supernatant and 5,385 pg/mL in neat synovial fluid.
Interpolated concentrations of native MIP5 in HT-29 cell extract based on a 300 μg/mL extract load and in human colon tissue extract based on a 500 μg/mL extract load.
The concentrations of MIP5 were measured in duplicate and interpolated from the MIP5 standard curve and corrected for sample dilution. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean MIP5 concentration was determined to be 1293 pg/mL in HT-29 cell extract and 685.5 pg/mL in colon tissue extract.
Serum from nine individual healthy human male donors was measured in duplicate.
Interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean MIP5 concentration was determined to be 7978 pg/mL with a range of 3361 – 13894 pg/mL.
MIP5 is increased in serum of patients with rheumatoid arthritis.
Serum was pooled from fifty apparently healthy donors and compared to serum from a patient who tested positive for rheumatoid factor (RF value 406.2 IU/mL). The concentrations of MIP5 were measured from serum samples in 3 different dilutions in duplicates and interpolated from the MIP5 standard curve. The interpolated values are plotted (mean +/- SD, n=3). The mean MIP5 concentration was determined to be 8753 pg/mL in healthy human serum and 26193 pg/mL in rheumatoid arthritis patient serum.
