Product details

Abcam's MIP5 Human ELISA (Enzyme-Linked Immunosorbent Assay) kit is an in vitro enzyme-linked immunosorbent assay for the quantitative measurement of Human MIP5 in serum, plasma, and cell culture supernatants.

This assay employs an antibody specific for Human MIP5 coated on a 96-well plate. Standards and samples are pipetted into the wells and MIP5 present in a sample is bound to the wells by the immobilized antibody. The wells are washed and biotinylated anti-Human MIP5 antibody is added. After washing away unbound biotinylated antibody, HRP-conjugated streptavidin is pipetted to the wells. The wells are again washed, a TMB substrate solution is added to the wells and color develops in proportion to the amount of MIP5 bound. The Stop Solution changes the color from blue to yellow, and the intensity of the color is measured at 450 nm.

Optimization may be required with urine samples.

Macrophage inflammatory protein 5 (MIP-5) also known as CCL15 is a chemokine involved in immune system function. It plays an important role in recruiting and activating certain immune cells such as monocytes and lymphocytes. Its molecular mass is approximately 13.5 kDa. MIP-5 is produced mainly by monocytes and macrophages and can be found in the liver and several other tissues. This protein mediates inflammation by guiding immune cells to sites of infection or injury where they can perform immune defense activities.
Biological function summary

MIP-5 is involved in the chemotactic movement of immune cells a process where cells move in response to chemical stimuli. It is not part of a protein complex but interacts dynamically with cell surface receptors to induce cellular responses. MIP-5 triggers cells to release further signaling molecules increasing the inflammatory response's effectiveness. This chemokine creates a highly coordinated immune response by initiating a signaling cascade that helps maintain immune surveillance and response to pathogens.

Pathways

MIP-5 plays a role in the chemokine signaling pathway where it interacts with specific receptors that lead to cellular migration and activation. It also fits into the cytokine-cytokine receptor interaction pathway influencing a range of cellular responses tied to immune function. MIP-5 shares functional properties with other chemokines like MCP-1 (monocyte chemoattractant protein-1) which similarly attract immune cells and mediate immune responses.

MIP-5 is linked to inflammatory diseases and certain types of cancer. Elevated levels of MIP-5 have been observed in rheumatoid arthritis where it contributes to joint inflammation. In these conditions MIP-5 might interact with other inflammatory molecules such as IL-6 and TNF-alpha suggesting its role as a mediator of chronic inflammation and malignancy processes.
