Product details

Abcam's MMP10 Human ELISA (Enzyme-Linked Immunosorbent Assay) kit is an in vitro enzyme-linked immunosorbent assay for the quantitative measurement of Human MMP10 pro and active forms in serum, plasma (Collect plasma using heparin as an anticoagulant. EDTA and Citrate are not recommended), and cell culture supernates.

This assay employs an antibody specific for Human MMP10 coated on a 96- well plate. Standards and samples are pipetted into the wells and MMP10 present in a sample is bound to the wells by the immobilized antibody. The wells are washed and biotinylated anti-Human MMP10 antibody is added. After washing away unbound biotinylated antibody, HRP-conjugated streptavidin is pipetted to the wells. The wells are again washed, a TMB substrate solution is added to the wells and color develops in proportion to the amount of MMP10 bound. The Stop Solution changes the color from blue to yellow, and the intensity of the color is measured at 450 nm.

Optimisation may be required with urine samples.

Supplementary information

MMP10 also known as stromelysin-2 is a member of the matrix metalloproteinase (MMP) family. This protein is involved in the degradation of components of the extracellular matrix. MMP10 weighs around 54 kDa in its native form. It gets expressed in various tissues including the skin lung and reproductive organs and plays a part in processes like tissue remodeling and repair. Unlike other MMPs MMP10 does not usually cleave collagen but breaks down proteins like elastin and gelatin. The Luminex MMP assay is a method used to study this protein's expression and regulation.
Biological function summary

MMP10 impacts the restructuring and turnover of the extracellular matrix in both normal and disease conditions. It does not work alone; it operates synergistically with other MMPs in a complex matrix framework. Within tissues MMP10 modifies the integrity of the extracellular environment affecting cell behaviors such as migration and invasion. It steps in during processes like wound healing and inflammation exemplifying its contribution to cellular repair mechanisms and their regulation.

Pathways

The activity of MMP10 intersects with the MAPK and TGF-beta signaling pathways. MMP10 facilitates these pathways by digesting extracellular matrix barriers which then influences cellular responses. MMP10's function is also linked with other metalloproteinases like MMP1 and MMP3 in the pathways. Its regulation and activation play an important role in maintaining cellular homeostasis and responding to environmental cues.

MMP10 is associated with conditions such as cancer and arthritis. In cancer its role in promoting extracellular matrix breakdown facilitates tumor cell invasion and metastasis. In arthritis MMP10 contributes to joint tissue destruction perpetuating the inflammatory cycle. This protein frequently collaborates with MMP13 in these diseases reflecting its significant part in disease progression and pathology.
