Sandwich ELISA - Human MMP9 ELISA Kit (AB246539)
Interpolated concentrations of native MMP9 in human lymph node tissue based on a 100 μg/mL extract load.
The concentrations of MMP9 were measured in duplicate and interpolated from the MMP9 standard curve and corrected for sample dilution. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean MMP9 concentration was determined to be 896.33 pg/mL in lymph node tissue extract.

Sandwich ELISA - Human MMP9 ELISA Kit (AB246539)
Interpolated concentrations of native MMP9 in human serum, plasma and cell culture supernatant samples.
The concentrations of MMP9 were measured in duplicates, interpolated from the MMP9 standard curves and corrected for sample dilution. Undiluted samples are as follows : serum 12.5%, plasma (heparin) 6.25%, saliva 6.25%, 1.5% PHA-M stimulated cell culture supernatant 5%, and unstimulated PBMC cell culture supernatant 100%. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean MMP9 concentration was determined to be 64.45 ng/mL in serum, 138.41 ng/mL in plasma (heparin) and 159.25 ng/mL in saliva, 167.62 ng/mL in 1.5% PHA-M stimulated cell culture supernatant, and 8.70 ng/mL in unstimulated PBMC cell culture supernatant.

Product details
Human MMP9 ELISA Kit ab246539 is a rapid single-wash 90-min Sandwich ELISA to measure Human MMP9 in cell culture extracts, cell culture supernatant, heparin plasma, saliva, serum, tissue extracts. This SimpleStep sensitivity is 22.17 pg/mL.
How the assay works
Human MMP9 SimpleStep ELISA®employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. See the SimpleStep ELISA® protocol summary in the image section for further details.
Assay Specificity
Our SimpleStep ELISA® kits use recombinant monoclonal antibodies rigorously validated to ensure the highest level of consistency and reproducibility, improved sensitivity and specificity and ease of scalability and security of supply.
Please refer to our protocol booklet for more details.

Biological function summary
Matrix metalloproteinase-9 profoundly influences tissue remodeling inflammation and angiogenesis. It does not form part of a larger protein complex but interacts closely with substrates in the extracellular matrix. MMP-9 plays important roles in processes such as embryogenesis reproduction and wound healing through the remodeling of surrounding tissues. The MMP-9 protein is often measured using assays like ELISA or analyzed via techniques such as Western blotting to determine its expression levels and activity in different biological contexts.
Pathways
MMP-9 participates in the matrix metalloproteinase pathway and the NF-kB signaling pathway. These pathways are essential in orchestrating processes such as tissue remodeling and inflammatory responses. MMP-9 acts alongside other matrix metalloproteinases including MMP-2 and is regulated by factors like cytokines growth factors and hormones ensuring the precision of extracellular matrix degradation during physiological processes.
