Interpolated concentrations of native MMP9 in human serum plasma and cell culture supernatant samples.

The concentrations of MMP9 were measured in duplicates interpolated from the MMP9 standard curves and corrected for sample dilution. Undiluted samples are as follows : serum 12.5% plasma (heparin) 6.25% saliva 6.25% 1.5% PHA-M stimulated cell culture supernatant 5% and unstimulated PBMC cell culture supernatant 100%. The interpolated dilution factor corrected values are plotted (mean +/- SD n=2). The mean MMP9 concentration was determined to be 64.45 ng/mL in serum 138.41 ng/mL in plasma (heparin) and 159.25 ng/mL in saliva 167.62 ng/mL in 1.5% PHA-M stimulated cell culture supernatant and 8.70 ng/mL in unstimulated PBMC cell culture supernatant.

The concentrations of MMP9 were measured in duplicate and interpolated from the MMP9 standard curve and corrected for sample dilution. The interpolated dilution factor corrected values are plotted (mean +/- SD n=2). The mean MMP9 concentration was determined to be 896.33 pg/mL in lymph node tissue extract.

How the assay works

Human MMP9 ELISA Kit, Chemiluminescent (ab327048) SimpleStep Ignite™ ELISA employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step during a 30 min only incubation, significantly reducing assay time. By utilizing a chemiluminscent detection method this improves the sensitivity and dynamic range when compared to traditional colorimetric ELISAs.

Matrix metalloproteinase that plays an essential role in local proteolysis of the extracellular matrix and in leukocyte migration (PubMed : 12879005, PubMed : 1480034, PubMed : 2551898). Could play a role in bone osteoclastic resorption (By similarity). Cleaves KiSS1 at a Gly-|-Leu bond (PubMed : 12879005). Cleaves NINJ1 to generate the Secreted ninjurin-1 form (PubMed : 32883094). Cleaves type IV and type V collagen into large C-terminal three quarter fragments and shorter N-terminal one quarter fragments (PubMed : 1480034). Degrades fibronectin but not laminin or Pz-peptide.
