Interpolated concentrations of native NDRG1 in human HeLa cell extract based on a 1,000 μg/mL extract load.

The concentrations of NDRG1 were measured in duplicate and interpolated from the NDRG1 standard curve and corrected for sample dilution. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean NDRG1 concentration was determined to be 13093.22 pg/mL in HeLa cell extract.

Product details

Human NDRG1 ELISA Kit (ab270214) is a single-wash 90 min sandwich ELISA designed for the quantitative measurement of NDRG1 protein in cell lysate. It uses our proprietary SimpleStep ELISA® technology. Quantitate Human NDRG1 with 75.2 pg/ml sensitivity.

SimpleStep ELISA® technology employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. See the SimpleStep ELISA® protocol summary in the image section for further details. Our SimpleStep ELISA® technology provides several benefits:

NDRG1 (N-myc downstream-regulated gene 1) also called DRG-1 and Cap43 is a 43 kDa member of the NDRG family of proteins. It is ubiquitously expressed, found in fibroblasts, endothelial cells, respiratory epithelium and prostate epithelium. NDRG1 is highly expressed in non-proliferating, differentiating tissue. Human NDRG1 is 394 amino acids in length and contains an NDGR domain (aa 286 - 316) plus three tandem 10 aa hydrophilic repeats (aa 339 - 368). There are three potential isoform variants; Two involve alternate start sites at Met82 and Met286, while a third shows a four aa substitution for aa 1 - 34. NDRG1 is a metastasis suppressor that works by blocking the angiogenesis signaling pathway and is a nickel- and calcium-inducible gene that plays important roles in the primary growth of malignant tumors, as well as in invasion and metastasis. NDRG1 shares 93.6%, 93.4%, 92.6%, and 99% sequence homology with mouse, rat, bovine, and monkey, respectively.

NDRG1 or N-Myc downstream-regulated gene 1 also known as Cap43 Drg-1 and RTP is a protein that plays key roles in various cellular processes. It has a molecular mass of approximately 43 kDa. NDRG1 is ubiquitously expressed with higher levels in the brain heart liver and kidney. The protein also exhibits a significant presence in cytoplasm but can translocate to the nucleus depending on the cellular context and stimuli.

Biological function summary

NDRG1 functions in cell growth regulation stress response and apoptosis influencing differentiation and potentially acting as a tumor suppressor. It does not directly form part of a larger protein complex but interacts with other proteins to mediate its effects. Its involvement in cellular homeostasis places it as a significant player in maintaining normal cell physiology.

Pathways

NDRG1 is involved in the hypoxia and stress response pathways regulating cellular responses to low oxygen and oxidative stress. It interacts with HIF-1α a critical regulator of the cellular response to hypoxia and also modulates PTEN and p53 proteins that are important for cell cycle control and apoptosis. These pathways and interactions reflect its function in cellular regulatory networks.

NDRG1 has associations with cancer and Charcot-Marie-Tooth disease type 4D. In cancer abnormal expression of NDRG1 has been linked to tumor progression and metastasis where it interacts with proteins like E-cadherin to influence cell adhesion and migration. In Charcot-Marie-Tooth disease mutations in the NDRG1 gene disrupt normal nerve function pointing to its importance in peripheral nervous system health.
