Human NEFL ELISA Kit (ab288171) is a single-wash 90 min sandwich ELISA designed for the quantitative measurement of Human NEFL protein in plasma and serum samples. It uses our proprietary SimpleStep ELISA® technology. Quantitate NEFL with 30.785 pg/mL sensitivity.
SimpleStep ELISA® technology employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. NEFL also known as neurofilament light polypeptide is a component of the cytoskeleton of neurons. It is one of the subunits of neurofilaments which also include medium and heavy chains. NEFL has a molecular mass of about 61.5 kDa. The protein is predominantly expressed in neurons throughout the central and peripheral nervous systems providing structural support and regulating axon diameter.
Neurofilament light polypeptide forms heteropolymers with other neurofilament proteins contributing to the assembly of stable neurofilament fibers. These fibers maintain neuronal integrity and proper conductance velocities of nerve signals. NEFL interacts closely with neurofilament medium and heavy chains playing a role in the structural framework of the neuron. This enables efficient transmission of electrical impulses along axons.
NEFL plays a significant role in the neurofilament pathway which is critical for neuronal cytoskeleton assembly and axonal transport. This pathway involves proteins such as NEFM (neurofilament medium) and NEFH (neurofilament heavy). NEFL's involvement in maintaining axonal caliber connects it to important signaling cascades like the axon guidance and neurotransmitter release pathways enhancing neuronal communication.
Defects in NEFL can lead to neurological conditions such as Charcot-Marie-Tooth disease type 2E and amyotrophic lateral sclerosis. Mutations or disruptions in NEFL impact axonal transport and neurofilament assembly contributing to neurodegeneration. NEFL is often studied alongside proteins like peripherin which can also exhibit mutations linked to similar neuropathies underlining its importance in neural health.

Interpolated concentrations of spiked human NEFL in serum, plasma (citrate), plasma (EDTA), and plasma (heparin). Recombinant NEFL was spiked into the following human samples and diluted in a 2-fold dilution series in Sample Diluent NS. Undiluted samples are 100% serum, plasma (citrate), plasma (EDTA), and plasma (heparin). The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2).
