Species reactivity was determined by measuring the protein in various samples, interpolating the protein concentrations from the human standard curve, and expressing the interpolated concentrations.
The mean Neurofilament H concentration was determined to be 5.301 μg/mg in human brain tissue extract, 3.386 μg/mg in mouse brain tissue extract, and 0.216 μg/mg in rat brain tissue extract.

Interpolated concentrations of native Neurofilament H in human SH-SY5Y cell extract based on a 250 µg/mL extract load and human brain tissue extract based on a 10 μg/mL extract load.
The concentrations of Neurofilament H were measured in duplicate and interpolated from the Neurofilament H standard curve and corrected for sample dilution. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean Neurofilament H concentration was determined to be 22.94 ng/mL in SH-SY5Y cell extract and 57.07 ng/mL in human brain tissue extract.

Human Neurofilament H ELISA kit (ab272199) is a single-wash 90 min sandwich ELISA designed for the quantitative measurement of Human Neurofilament H protein in human cell and tissue extract samples. It uses our proprietary SimpleStep ELISA® technology. Quantitate Human Neurofilament H with 1.41 pg/mL sensitivity.
SimpleStep ELISA® technology employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. Neurofilament H, also known as 200 kDa Neurofilament Protein, together with Neurofilament L and M are three intermediate filament proteins of neurofilaments. Neurofilament H is involved in the maintenance of neuronal caliber and may play a role in intracellular transport. Neurofilament H is used as a neuronal damage biomarker and mutations to the gene have been associated with susceptibility to amyotrophic lateral sclerosis. The human Neurofilament H amino acids sequence is 75% identical to mouse and rat Neurofilament H sequence.

Biological function summary
This polypeptide participates in the formation of neurofilament a complex structure that includes other neurofilament proteins like the neurofilament light (NF-L) and neurofilament medium (NF-M). These proteins form a stable scaffold within the axon aiding in neuronal growth and repair. The phosphorylation of NF-H modulates its interactions within the neurofilament network influencing axonal transport processes and dynamics which are vital for neuron function and survival.
Pathways
Neurofilament heavy polypeptide is integral to the axonal transport pathway important in conveying molecules between the neuronal cell body and the synaptic terminals. Through its interactions with microtubules and motor proteins NF-H facilitates the movement of organelles and neurotransmitters. It has significant associations with proteins involved in the ubiquitin-proteasome system reflecting its role in maintaining neuronal homeostasis and proteostasis essential in neuronal development and function.
