Interpolated concentrations of spike Noggin in human serum, plasma samples.
The concentrations of Noggin were measured in duplicates, interpolated from the Noggin standard curves and corrected for sample dilution. Undiluted samples are as follows : serum 50%, plasma - citrate 50%, plasma - EDTA 50%. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2).

Native Noggin detected in serum from ten individual healthy human female donors was measured in duplicate.
Interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean Noggin concentration was determined to be 99 pg/mL with a range of 30 – 260 pg/mL.

Human Noggin ELISA Kit (ab270218) is a single-wash 90 min sandwich ELISA designed for the quantitative measurement of Noggin protein in serum and plasma. It uses our proprietary SimpleStep ELISA® technology. Quantitate Human Noggin with 11 pg/ml sensitivity.
SimpleStep ELISA® technology employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. Noggin, also known as NOG, is a signaling molecule that regulates the function of bone morphogenic protein. It is known to play a key role in embryonic development, learning, and bone homeostasis. Dysregulation of this protein leads to multiple deformities. The immunogen from which the antibodies in this kit were raised represent the full-length mature protein. This protein is homologous with many species, so cross-reactivity is expected.

Biological function summary
Noggin functions as an essential regulator in embryonic development sculpting tissues by inhibiting BMP activity. It does not work alone but functions as part of a complex regulatory network involving other BMP antagonists. By inhibiting BMPs Noggin plays an important role in neural tube closure limb formation and somite development. Noggin's activities help modulate the balance between proliferation and differentiation in these contexts allowing for proper structural formation during embryogenesis.
Pathways
Noggin operates mainly within the TGF-beta/BMP signaling pathway serving as a critical modulator. Inhibiting BMP through Noggin affects downstream targets connecting it to other signaling proteins like SMADs which mediate transcriptional responses. This control enables Noggin to indirectly regulate Wnt signaling as well showing its impact on various developmental stages and processes. Its influence on these pathways indicates Noggin’s importance in maintaining cellular homeostasis during growth and morphogenesis.
