Abcam's Notch-1 (TAN-1) Human ELISA (Enzyme-Linked Immunosorbent Assay) kit is an in vitro enzyme-linked immunosorbent assay designed for the quantitative measurement of Human Notch-1 (TAN-1) in serum, plasma and cell culture supernatants.

This assay employs an antibody specific for Human Notch-1(TAN-1) coated on a 96-well plate. Standards and samples are pipetted into the wells and Notch-1 present in a sample is bound to the wells by the immobilized antibody. The wells are washed and biotinylated Anti-Human Notch-1 antibody is added. After washing away unbound biotinylated antibody, HRP-conjugated streptavidin is pipetted to the wells. The wells are again washed, a TMB substrate solution is added to the wells and color develops in proportion to the amount of Notch-1 bound. The Stop Solution changes the color from blue to yellow, and the intensity of the color is measured at 450 nm.

Optimization may be required with urine samples.

This kit can detect extracellular domain of NOTCH1.

Notch-1 also known as TAN-1 is a transmembrane receptor protein with a molecular mass of approximately 300 kDa. It is part of the Notch family and is extensively expressed in various tissues including the brain heart and immune cells. The receptor plays an important role in cell-fate determination by mediating communication between adjacent cells through its interaction with notch ligands like Delta and Jagged.
Biological function summary

Notch-1 directly influences cell differentiation proliferation and apoptosis by participating in the Notch signaling pathway. It is part of a complex with Notch ligands and the γ-secretase enzyme which facilitates its cleavage and activation. This signaling assists in controlling the balance between cell types in the developing organism and adult tissue homeostasis making it important for proper function and organism development.

Pathways

Notch-1 is involved in regulating the Notch signaling pathway which impacts gene expression once the intracellular domain translocates to the nucleus. Additionally it interacts with proteins such as Mastermind-like proteins to activate transcription of target genes. Notch-1 also intersects with the Wnt signaling pathway suggesting crosstalk between these important pathways in maintaining cell fate decisions and tissue development.

Notch-1 has connections to T-cell acute lymphoblastic leukemia as mutations in Notch-1 often lead to uncontrolled cell proliferation. The receptor is also linked to breast cancer where abnormal Notch-1 signaling encourages tumor progression. In these disorders Notch-1 interacts with proteins such as NF-kB which influences cancer cell survival and proliferation making Notch-1 a significant target for therapeutic interventions.

Upon ligand activation through the released notch intracellular domain (NICD) it forms a transcriptional activator complex with RBPJ/RBPSUH and activates genes of the enhancer of split locus. Affects the implementation of differentiation, proliferation and apoptotic programs. Involved in angiogenesis; negatively regulates endothelial cell proliferation and migration and angiogenic sprouting. Involved in the maturation of both CD4(+) and CD8(+) cells in the thymus. Important for follicular differentiation and possibly cell fate selection within the follicle. During cerebellar development, functions as a receptor for neuronal DNER and is involved in the differentiation of Bergmann glia. Represses neuronal and myogenic differentiation. May play an essential role in postimplantation development, probably in some aspect of cell specification and/or differentiation.
