Interpolated concentrations of human NOTUM in HepG2 Extract, Placenta Extract. Interpolated concentration of native NOTUM was measured in duplicate at different sample concentrations. Undiluted samples are as follows : HepG2 Extract 250 ug/mL and Placenta Extract 500 ug/mL. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). Sample dilutions are made in 1X Cell Extraction Buffer PTR.

Human NOTUM SimpleStep ELISA® kit is a single-wash 90 min sandwich ELISA designed for the quantitative measurement of NOTUM protein in Cell Lysate, Tissue extracts. Quantitate Human NOTUM with 17.047 pg/ml sensitivity.
SimpleStep ELISA® technology employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. See the SimpleStep ELISA® protocol summary in the image section for further details. Our SimpleStep ELISA® technology provides several benefits:
-Single-wash protocol reduces assay time to 90 minutes or less

Biological function summary
NOTUM inhibits the Wnt signaling pathway by cleaving the lipid moiety attached to Wnt proteins making it inactive. NOTUM acts independently and is not part of larger protein complexes. This regulation of Wnt signaling is important for maintaining cellular processes such as cell proliferation differentiation and migration especially during development and tissue homeostasis.
Pathways
NOTUM primarily influences the Wnt signaling pathway. This pathway is significant for embryonic development and adult homeostasis. By inactivating Wnt proteins NOTUM indirectly affects other pathway components like β-catenin which is an important mediator in the Wnt pathway. NOTUM ensures the balance of Wnt signaling preventing overactivation that could lead to abnormal cell growth.
