May promote proliferation of pancreatic cancer cells by favoring the transition from the S to G2/M phase. Human OLFM4 ELISA Kit (ab267805) is a single-wash 90 min sandwich ELISA designed for the quantitative measurement of OLFM4 protein in serum, cit plasma, tissue extracts, and cell culture media. It uses our proprietary SimpleStep ELISA® technology. Quantitate Human OLFM4 with 315.7 pg/ml sensitivity.
SimpleStep ELISA® technology employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. Olfactomedin-4 (OLFM4), formerly known as GW112 or hGC-1, is a member of the olfactomedin gene family and predominantly expressed in bone marrow, small intestine, colon, and prostrate. Studies have revealed OLFM4 to play important roles in several cellular functions such as neurogenesis, cell adhesion, cell-cycle regulation, and tumorigenesis. Moreover, studies have suggested OLFM4 to serve as a robust biomarker in studying stem cells and cancer in small intestines, stomach, and colon.

The OLFM4 protein also known as Olfactomedin 4 or GW112 has a molecular weight of around 57 kDa. It is a glycoprotein expressed mainly in human and some non-human primate tissues including the prostate and gastrointestinal tract. OLFM4 is a secreted protein found in various cells including those of the bone marrow and esophagus. It plays a role in innate immunity and is a part of the olfactomedin family which is involved in cell adhesion and migration processes.
OLFM4 functions in inhibiting bacterial growth by interfering with bacterial adhesion and survival. It is not part of a larger protein complex but interacts with cell surface proteins to exert its effects. OLFM4 acts in the regulation of cell differentiation and growth playing a significant part in maintaining the stem cell milieu in the intestine.
OLFM4 participates in cellular defense mechanisms primarily against microbial pathogens by involving in the activation of immune response pathways. It is associated with pathways such as the NF-kB signaling and Janus kinase (JAK) pathways. Through these pathways OLFM4 might interact with cytokines and other inflammatory mediators or modulators.
OLFM4 relates to cancer particularly gastric and colorectal cancer where its expression levels serve as potential biomarkers for disease progression. Additionally it associates with inflammatory bowel diseases impacting the mucosal immune response. In cancer OLFM4's interaction with proteins like MMP isoforms (matrix metalloproteinases) can influence tumor metastasis and invasiveness.

The OLFM4 standard curve was prepared as described in Section 10. Raw data values are shown in the table. Background-subtracted data values (mean +/- SD) are graphed.
Interpolated concentrations of native OLFM4 in human serum and plasma samples.
The concentrations of OLFM4 were measured in duplicates, interpolated from the OLFM4 standard curves and corrected for sample dilution. Undiluted samples are as follows: serum 100% and plasma (citrate) 100%. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean OLFM4 concentration was determined to be 4,710 pg/mL in serum and 4,029 pg/mL in plasma (citrate).
Interpolated concentrations of native OLFM4 in human colon tissue extract based on a 400 μg/mL extract load.
The concentrations of OLFM4 were measured in duplicate and interpolated from the OLFM4 standard curve and corrected for sample dilution. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean OLFM4 concentration was determined to be 48,973 pg/mL in colon tissue extract.
