Oligodendrocyte myelin glycoprotein (OMGP) also known as oligodendrocyte myelin marker has a molecular weight of approximately 50 kDa. It is a glycoprotein primarily found in the myelin of the central nervous system (CNS) notably in the white matter. OMGP appears as an element on the surface of oligodendrocytes and contributes significantly to the stability and function of the myelin sheath.
Biological function summary

OMGP plays an essential role in the CNS where it interacts with other structural components to maintain myelin integrity. OMGP is not a component of a larger complex but works closely with other myelin proteins such as myelin basic protein. It also inhibits neurite outgrowth helping regulate neural plasticity and potentially preventing improper nerve regeneration in mature central nervous systems.

Pathways

Research recognized OMGP as a contributor in the Nogo receptor (NgR) signaling pathway influencing the inhibition of axonal growth. Within this pathway OMGP associates with other proteins like Nogo-A and the receptor NgR1 regulating neuronal growth by binding to these proteins and suppressing neurite outgrowth. This interaction is important in maintaining the structural arrangement of the CNS and modulating its repair mechanisms.

OMGP has connections with multiple sclerosis (MS) and some neurodegenerative diseases. In MS changes in OMGP expression correlate with demyelination and faulty nerve function. Moreover OMGP interrelates with Nogo-A another protein present in damaged myelin which may influence disease progression by hindering nerve repair. ALS (Amyotrophic Lateral Sclerosis) research also considers OMGP's role in neurodegeneration suggesting a broader impact on CNS pathologies.
