Interpolated concentrations of native P-Selectin in human serum and plasma (EDTA).
The concentrations of P-Selectin were measured in duplicates, interpolated from the target standard curves and corrected for sample dilution. Undiluted samples are as follows : serum 10% and plasma (EDTA) 10%. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean target concentration was determined to be 65.6 ng/mL in serum and 54.3 ng/mL in plasma (EDTA).

Serum from eight individual healthy P-Selectin gender donors was measured in duplicate.
Interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean P-Selectin concentration was determined to be 76.7 ng/mL with a range of 50.1 – 163.8 ng/mL.

Human P-Selectin ELISA kit (ab272200) is a single-wash 90 min sandwich ELISA designed for the quantitative measurement of P-Selectin protein in serum, plasma and cell culture supernatant. It uses our proprietary SimpleStep ELISA® technology. Quantitate Human P-Selectin with 89 pg/mL sensitivity.
SimpleStep ELISA® technology employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. P-selectin, also known as CD62, Granule membrane protein 140, Leukocyte-endothelial cell adhesion molecule 3, or platelet activation dependent granule-external membrane protein, is a cell adhesion molecule (CAM) encoded by the SELP gene. P-selectin is Ca2+ dependent cell-surface glycoprotein that functions in the migration of lymphocytes into tissues during inflammation. P-selectin expressed by megakaryocytes and endothelial cells, where it is stored in Weibel-Palade bodies and alpha-granules. In the presence of agonists such as thrombin, P-selectin is translocated to the plasma membrane from granules. P-selectin expression is also induced by inflammatory agents such as; TNF-a, LPS, and IL-4. The ligand for P-selectin is P-Selectin glycoprotein ligand-1 (PSGL-1), which is primarily expressed on all leukocytes.

Biological function summary
P-selectin is critical in the recruitment of leukocytes to sites of tissue injury and infection. It facilitates the adhesion of leukocytes to the vascular endothelium by binding to its ligand PSGL-1 on the leukocyte surface. This interaction supports the transition of leukocytes from the circulation to the site of inflammation. P-selectin is also involved in thrombus formation as it contributes to the interactions between platelets leukocytes and endothelium in the vascular wall. It acts as part of a dynamic complex in the regulation of the innate immune response and hemostasis.
Pathways
CD62P plays an important role in both the inflammatory and coagulation pathways. It is an important mediator in the leukocyte adhesion cascade influencing leukocyte tethering and rolling on activated endothelium. The interaction of P-selectin with PSGL-1 initiates the signaling pathways that lead to firm adhesion and extravasation of leukocytes. In the coagulation pathway P-selectin enhances interactions between platelets and leukocytes which support thrombus formation. Its regulation and activity are tightly linked with other proteins such as E-selectin and L-selectin in coordinating vascular and systemic inflammatory responses.
