Abcam's Human Amylase (pancreatic/salivary) in vitro ELISA (Enzyme-Linked Immunosorbent Assay) kit is designed for the quantitative measurement of Human pancreatic and salivary amylase in plasma, serum, urine, milk, saliva samples.

A Pancreatic and Salivary Amylase specific antibody has been precoated onto 96-well plates and blocked. Standards or test samples are added to the wells and subsequently a Pancreatic/Salivary Amylase specific biotinylated detection antibody is added and then followed by washing with wash buffer. Streptavidin-Peroxidase Conjugate is added and unbound conjugates are washed away with wash buffer. TMB is then used to visualize Streptavidin-Peroxidase enzymatic reaction. TMB is catalyzed by Streptavidin-Peroxidase to produce a blue color product that changes into yellow after adding acidic stop solution. The density of yellow coloration is directly proportional to the amount of Pancreatic Amylase captured in plate.

The entire kit may be stored at -20°C for long term storage before reconstitution - Avoid repeated freeze-thaw cycles.

Pancreatic amylase also known as alpha-amylase or amylopsin is an enzyme with a molecular mass of approximately 55 kDa. This enzyme is expressed mainly in the pancreas and is secreted into the small intestine. Its primary mechanical function involves hydrolyzing starch into smaller carbohydrates such as maltose and dextrin. Pancreatic amylase activity is essential for the efficient digestion of dietary starch aiding in the breakdown into sugars that the body can easily absorb and utilize.
Biological function summary

The enzyme facilitates the digestive process by catalyzing the cleavage of 14-glycosidic bonds in polysaccharides. Pancreatic amylase operates independently and not as part of a larger protein complex. Its function exclusively converts complex carbohydrates into simpler absorbable sugars.
