PAX4 also known as paired box 4 is a protein encoded by the PAX4 gene. This transcription factor has a molecular mass of around 36 kDa. It plays a significant role in the regulation of pancreatic islet development and insulin gene expression. PAX4 is expressed mainly in pancreatic beta cells. Its expression is important for the proper differentiation of these cells from precursor cells and is pivotal during embryogenesis in the formation of pancreatic endocrine cells.
Biological function summary

PAX4 influences cellular activities critical for the development of insulin-producing cells in the pancreas. As a transcription factor it does not work in isolation but interacts with other proteins taking part in complexes that drive gene expression changes. It represses the expression of genes like glucagon ensuring the adequate development and function of insulin-secreting cells. Therefore PAX4 ensures a delicate balance in glucose homeostasis.

Pathways

PAX4 plays a significant part in the insulin signaling pathway and is essential in pathways affecting glucose metabolism. It interacts with RXR and PDX1 which are key proteins in the regulation of insulin and glucose metabolism genes. By controlling these pathways PAX4 contributes to maintaining normal glucose levels and effective insulin function which is critical for energy balance in the body.
