Product details
Human PBK (PDZ Binding Kinase) ELISA kit (ab184866) is a single-wash 90 min sandwich ELISA designed for the quantitative measurement of PBK protein in cell and tissue extracts. It uses our proprietary SimpleStep ELISA® technology. Quantitate human PDZ Binding Kinase with 133 pg/mL sensitivity.
SimpleStep ELISA® technology employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. See the SimpleStep ELISA® protocol summary in the image section for further details. Our SimpleStep ELISA® technology provides several benefits:
-Single-wash protocol reduces assay time to 90 minutes or less
-High sensitivity, specificity and reproducibility from superior antibodies
-Fully validated in biological samples
-96-wells plate breakable into 12 x 8 wells strips
A 384-well SimpleStep ELISA® microplate (ab203359) is available to use as an alternative to the 96-well microplate provided with SimpeStep ELISA® kits.
PDZ-binding kinase (also known PBK or Lymphocyte-activated killer T-cell-originated protein kinase) is a Serine/Threonine protein kinase in the MAP kinase kinase subfamily. PBK phosphorylates MAP kinase p38. PBK itself is phosphorylated and appears to be active only during the mitotic phase of the cell cycle. When phosphorylated PBK may interact with TP53. PBK may play a role in the activation of lymphoid cells and is found to expressed in many highly proliferative cell lines.
The standard is affinity purified.

Biological function summary
PBK acts as part of cellular proliferation and differentiation processes. It can form complexes with other proteins enhancing its function and stability. The kinase significantly contributes to cell growth mainly in rapidly dividing cells. PBK influences processes like spermatogenesis in testes and placental development. PBK stabilizes certain cyclins and links with proteins such as p38 MAP kinase showing a role in stress response pathways.
Pathways
PBK integrates into the mitogen-activated protein kinase (MAPK) pathway and interacts with the tumor progression pathway. These pathways are critical for transmitting signals from the surface to the nucleus affecting cell fate decisions. PBK connects with proteins like JNK and extracellular signal-regulated kinase (ERK) displaying its importance in transmitting stress and growth signals. These interactions place PBK as a node linking growth factors and extracellular signals to cellular responses.
