Interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean PD-1 concentration was determined to be 86.56 pg/mL with a range of 47.82 – 210.99 pg/mL.

Interpolated concentrations of native PD-1 in human serum, plasma samples.
The concentrations of PD-1 were measured in duplicates, interpolated from the PD-1 standard curves and corrected for sample dilution. Undiluted samples are as follows : serum neat, plasma (citrate) neat, plasma (EDTA) neat, plasma (heparin) neat. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean PD-1 concentration was determined to be 119.1 pg/mL in serum, 88.1 pg/mL in plasma (citrate) and 85.9 pg/mL in plasma (EDTA), and 92.8 pg/mL in plasma (heparin).

Interpolated concentrations of native PD-1 in human Molt-4 cell extract based on a 1000 ug/mL extract load and Thymus tissue extract samples based on a 500 μg/mL extract load.
The concentrations of PD-1 were measured in duplicate and interpolated from the PD-1 standard curve and corrected for sample dilution. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean PD-1 concentration was determined to be 636.9 pg/mL in Molt-4 cell extract and 566.5 pg/mL in Thymus tissue extract.

How the assay works

1. Mix: add samples/standards to the wells together with the capture and detector antibody cocktail. Incubate 1 hr at room temperature

3. Add TMB development solution - incubate for 10 min

Biological function summary
PD1 serves as an inhibitory receptor acting as a checkpoint in the immune system. It becomes part of an immune-suppressive complex when it binds with its ligands PD-L1 or PD-L2 which are expressed on various cell types including some tumor cells. This interaction suppresses the proliferation of T cells and cytokine production contributing to immune homeostasis. By controlling T cell activity PD1 limits autoimmunity but can also reduce the immune system's capability to attack cancer cells.
Pathways
PD1 functions in the immune checkpoint pathway a critical regulatory circuit in immune regulation. The engagement of PD1 with its ligands initiates a cascade that inhibits the function and proliferation of T cells through downstream SHP-2 phosphatase activity. This pathway frequently involves other regulatory proteins like CTLA-4 and is an important mechanism by which the body modulates immune responses. Related pathways often intersect with those involving T cell receptor signaling and contribute to the overall modulation of immune activity.
