Human peripheral blood cells (1x106 cells/mL) were cultured in RPMI media supplemented with 10% fetal calf serum, 100 U/mL penicillin, and 100 µg/mL streptomycin sulfate.
Cells were cultured unstimulated or stimulated with 10 μg/mL PHA. Conditioned media was harvested after 48 hours aliquoted and assayed for endogenous PDGF-BB levels.

How the assay works

This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. Our SimpleStep ELISA® kits use recombinant monoclonal antibodies rigorously validated to ensure the highest level of consistency and reproducibility, improved sensitivity and specificity and ease of scalability and security of supply.
This immunoassay is calibrated against a highly purified human PDGF BB. The NIBSC/WHO unclassified purified human PDGF BB preparation 94/728 was evaluated in this kit.
The dose response curve of the unclassified standard PDGF BB parallels the SimpleStep standard curve.
To convert sample values obtained with the SimpleStep Human PDGF BB kit to approximate NIBSC IU/mL units, use the equation below.
NIBSC (94/728) approximate value (IU/mL) = 0.0114 x SimpleStep Human PDGF BB value (pg/mL).

Biological function summary
PDGF-B influences cellular signaling processes and facilitates interactions that are a part of the PDGF receptor system. PDGF-B can form a homo-dimerization commonly known as the PDGF-BB dimer which is an active form. When PDGF-B binds to its receptor specifically the PDGF receptor β (PDGFR-β) it triggers pathways that control various cellular activities including growth and survival.
