Serum of ten individual healthy human donors was measured in duplicate. Interpolated dilution factor corrected values (to neat sample) are plotted (mean +/- SD, n=2). The mean PDGF-BB concentration was determined to be 145 pg/mL with a range of 57.9 – 286.5 pg/mL.

Human peripheral blood cells (1x106 cells/mL) were cultured in RPMI media supplemented with 10% fetal calf serum, 100 U/mL penicillin, and 100 µg/mL streptomycin sulfate. Cells were cultured and treated in the absence and presence of 10 µg/mL PHA. Conditioned media was harvested after 48 hours aliquoted and assayed for endogenous PDGF-BB levels.

Elevated levels PDGF-BB were measured in duplicate in the serum of six individual Rheumatoid Arthritis donors. Interpolated dilution factor corrected values (to neat sample) are plotted (mean +/- SD, n=2). The mean PDGF-BB concentration was determined to be 8,766.1 pg/mL with a range 6193.9 – 11,336.1 pg/mL.

How the assay works

Human PDGF BB ELISA Kit, Chemiluminescent (ab325904) SimpleStep Ignite™ ELISA employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step during a 30 min only incubation, significantly reducing assay time. By utilizing a chemiluminscent detection method this improves the sensitivity and dynamic range when compared to traditional colorimetric ELISAs.

Target data

Growth factor that plays an essential role in the regulation of embryonic development, cell proliferation, cell migration, survival and chemotaxis. Potent mitogen for cells of mesenchymal origin (PubMed : 26599395). Required for normal proliferation and recruitment of pericytes and vascular smooth muscle cells in the central nervous system, skin, lung, heart and placenta. Required for normal blood vessel development, and for normal development of kidney glomeruli. Plays an important role in wound healing. Signaling is modulated by the formation of heterodimers with PDGFA (By similarity).
