Interpolated concentrations of native Pentraxin 3 in human HUVEC Extract sample based on a 200 µg/mL extract load.
The concentrations of Pentraxin 3 were measured in duplicate and interpolated from the Pentraxin 3 standard curve and corrected for sample dilution. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean Pentraxin 3 concentration was determined to be 1,368 pg/mL.

Interpolated concentrations of native Pentraxin 3 in human plasma, saliva and cell culture supernatant samples.
The concentrations of Pentraxin 3 were measured in duplicates, interpolated from the Pentraxin 3 standard curves and corrected for sample dilution. Undiluted samples are as follows : Plasma (heparin) 25%, plasma (EDTA) 25%, saliva 10%, and HUVEC cell culture supernatant 100%. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean Pentraxin 3 concentration was determined to be 1,056 pg/mL in plasma (heparin), 1,329 og/mL in plasma (EDTA), 962 pg/mL in saliva and 334 pg/mL in HUVEC cell culture supernatant.

Human Pentraxin 3 / PTX3 ELISA Kit (ab214570) is a single-wash 90 min sandwich ELISA designed for the quantitative measurement of Pentraxin 3 / PTX3 protein in cell culture extracts, cell culture supernatant, cit plasma, edta plasma, hep plasma, serum, tissue extracts, and saliva. It uses our proprietary SimpleStep ELISA® technology. Quantitate Human Pentraxin 3 / PTX3 with 3.4 pg/ml sensitivity.
SimpleStep ELISA® technology employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. Pentraxin 3 (also known as pentraxin-related protein PTX3 or TSG-14) is a secreted protein with a role in innate immune response. It is secreted by many cell types during infectious situations. The standard protein provided in this kit corresponds to the C-terminal Pentaxin domain of Pentraxin 3.

Biological function summary
The role of PTX3 extends beyond simple pathogen recognition. It is involved in the regulation of inflammation and tissue homeostasis. As part of the pentraxin superfamily PTX3 plays a role in the formation of complexes with other molecules such as complement components. These interactions modulate the complement cascade an essential part of the innate immune response and influence the resolution of inflammation. PTX3 also interacts with extracellular matrix components stabilizing tissue structure during immune response.
Pathways
PTX3 integrates into vital immune signaling networks. It actively participates in the complement pathway where it regulates the activity of C1q and C3 key complement proteins. Additionally PTX3 connects with the Toll-like receptor (TLR) signaling pathway. This interaction helps modulate the body's response to microbial infections by influencing the activity of TLR4 among other related proteins which are fundamental in recognizing pathogen-associated molecular patterns.
