Abcam's Perforin Human in vitro ELISA Set is designed for the quantitative measurement of Perforin in cell culture supernatants and buffered solutions.
The Perforin ELISA Set includes both the capture and the detection antibodies as well as Horse-Radish peroxidase conjugated Streptavidin (HRP-Strep) and TMB which are the reagents used in the colour development step. This ELISA set is intended to measure Perforin in cell -culture supernatants, as well as in serum samples*. The enclosed procedures provide general guidelines for ELISA preparation. This procedure may need some optimization depending on the plastic, matrix, standard range.
*Special note for serum & plasma quantification: high and non linear detection level is found in human serum and plasma samples. The high signal appears not dependant to the antibody pairs but probably to the matrix and/ or perforin interaction with other molecules. Consequently serum and plasma quantification is under the scientist responsibility and specific investigations. Remark: no detection signal was found in fetal calf serum, rat, mouse and horse serum

Biological function summary
Perforin helps the immune system respond effectively against infected or malignant cells. It works in conjunction with granzymes forming a complex that perforates target cell membranes allowing granzymes to enter and induce apoptosis. Perforin's role in immune surveillance prevents the propagation of harmful cells within the body. Deficiency or malfunction in perforin can lead to compromised immune function or immune cell dysregulation.
Pathways
Perforin integrates into the cytotoxic T lymphocyte (CTL) and NK cell-mediated cytotoxicity pathways. These pathways include critical interactions with proteins like granzymes and Fas ligand (FasL) which orchestrate the apoptosis of abnormal cells. Through these pathways perforin ensures the body's ability to identify and eliminate threats. The granule exocytosis pathway in particular heavily depends on perforin's activity to execute effective immune responses.
