Sandwich ELISA - Human Polymeric immunoglobulin receptor ELISA Kit (PIGR) (AB282302)
Interpolated concentrations of native Polymeric immunoglobulin receptor in human stomach tissue extract based on a 500 μg/mL extract load.
The concentrations of Polymeric immunoglobulin receptor were measured in duplicate and interpolated from the Polymeric immunoglobulin receptor standard curve and corrected for sample dilution. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean Polymeric immunoglobulin receptor concentration was determined to be 11.18 ng/mL in stomach tissue extract.

Sandwich ELISA - Human Polymeric immunoglobulin receptor ELISA Kit (PIGR) (AB282302)
Interpolated concentrations of native Polymeric immunoglobulin receptor in human serum and plasma samples.
The concentrations of Polymeric immunoglobulin receptor were measured in duplicates, interpolated from the Polymeric immunoglobulin receptor standard curves and corrected for sample dilution. Undiluted samples are as follows : serum 1.25%, plasma (citrate) 1.25%, plasma (EDTA) 1.25% and plasma (heparin) 1.25%. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean Polymeric immunoglobulin receptor concentration was determined to be 613.22 ng/mL in serum, 713.68 ng/mL in plasma (citrate), 789.90 ng/mL in plasma (EDTA), and 666.13 ng/mL in plasma (heparin).

Product details
Human Polymeric immunoglobulin receptor ELISA Kit (PIGR) ab282302 is a rapid single-wash 90-min Sandwich ELISA to measure Human Polymeric immunoglobulin receptor (PIGR)in cell culture media, citrate plasma, EDTA plasma, heparin plasma, serum. This SimpleStep sensitivity is 29.16 pg/mL.
How the assay works
Human Polymeric immunoglobulin receptor (PIGR)SimpleStep ELISA®employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. See the SimpleStep ELISA® protocol summary in the image section for further details.
Assay Specificity
Our SimpleStep ELISA® kits use recombinant monoclonal antibodies rigorously validated to ensure the highest level of consistency and reproducibility, improved sensitivity and specificity and ease of scalability and security of supply.
Please refer to our protocol booklet for more details.

Human Polymeric immunoglobulin receptor (PIGR) mediates selective transcytosis of polymeric IgA and IgM across mucosal epithelial cells. PIGR binds polymeric IgA and IgM at the basolateral surface of epithelial cells and the complex is then transported across the cell to be secreted at the apical surface. During this process, a cleavage occurs that separates the PIGR extracellular (known as the secretory component) from the transmembrane segment. This kit was designed to target the extracellular region of PIGR. The extracellular region of PIGR shares 65%, 64%, 68% and, 88% sequence homology with mouse, rat, cow, and monkey, respectively.

Biological function summary
The polymeric immunoglobulin receptor plays an essential role in mucosal immunity by transporting IgA and IgM antibodies to mucosal surfaces. The PIGR is a part of a complex that includes the secretory component which protects immunoglobulins from proteolytic degradation. This mechanism is fundamental for safeguarding mucosal surfaces while allowing the immune system to respond swiftly to pathogens.
Pathways
The interaction of PIGR with polymeric immunoglobulins is an important event in the mucosal immune pathway. This pathway is important in maintaining mucosal homeostasis and pathogen defense. PIGR works closely with immunoglobulin proteins like IgA and IgM enabling their rapid and efficient transport across epithelial barriers. This ensures that the immune system rapidly addresses potential threats at mucosal sites.
