The Human Proteinase 3 Autoantibody ELISA (ab284018) Kit is designed for the quantitative determination of autoimmune response (IgG) to a target antigen (PR3). The kit detects autoantibodies in plasma and serum samples.
This assay employs a quantitative sandwich enzyme immunoassay technique that measures autoantibodies (anti-PR3 IgG) in approximately 4 hours. A PR3 antigen has been pre-coated onto a 96-well microplate with removable strips. An autoantibody specific for PR3 in standards and samples is sandwiched by the immobilized antigen and a biotinylated polyclonal antibody specific for human IgG, which is recognized by a streptavidin-peroxidase (SP) conjugate. All unbound material is washed away and a peroxidase enzyme substrate is added. The color development is stopped and the intensity of the color is measured.

Pathways
Proteinase 3 plays an important role within the serine protease and inflammatory pathways. It interacts with other proteins such as neutrophil elastase with which it shares a pathway to modulate immune responses. PR3 also partners with cathepsin G contributing to processes such as pathogen clearance and tissue remodeling providing a coordinated response during infection and injury. These interactions show PR3's involvement in regulating the balance between inflammation and tissue repair.
