Interpolated concentrations of native PRAME in human K562 cell extract based on a 100 µg/mL extract load and testis tissue extract based on a 180 μg/mL extract load.
The concentrations of PRAME were measured in duplicate and interpolated from the PRAME standard curve and corrected for sample dilution. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean PRAME concentration was determined to be 14.70 ng/mL in K562 cell extract and 2.359 ng/mL in testis tissue extract.

How the assay works
Human PRAME SimpleStep ELISA®employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. 1. Mix: add samples/standards to the wells together with the capture and detector antibody cocktail. Incubate 1 hr at room temperature
2. Wash
3. Add TMB development solution - incubate for 10 min
4. Add Stop solution
5. Read the results on a plate reader at 450 nm

Biological function summary
PRAME acts as a repressor of retinoic acid signaling a process important for cell differentiation and growth. It does not directly form part of a complex but interacts with components involved in retinoic acid pathways. By binding to retinoic acid receptor complexes PRAME prevents activation of genes involved in cell cycle arrest and apoptosis contributing to unchecked cellular proliferation seen in certain cancers.
Pathways
PRAME participates in the retinoic acid and various oncogenic signaling processes. It influences the retinoic acid pathway by interfering with the retinoic acid receptor (RAR) signaling. This interference with key proteins like RAR disrupts the normal regulatory processes that typically inhibit cancer progression. PRAME's modulation of these pathways highlights its role in promoting tumor growth and survival.
