Interpolated concentrations of Prekallikrein 1B levels in individual healthy donors.
Serum from 10 healthy male donors was measured in duplicate. The mean Plasma Kallikrein 1B concentration was determined to be 112.6 μg/mL with a range of 80.8 – 206.4 μg/mL in male donors.

How the assay works
Human Prekallikrein 1B SimpleStep ELISA®employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. 1. Mix: add samples/standards to the wells together with the capture and detector antibody cocktail. Incubate 1 hr at room temperature
2. Wash
3. Add TMB development solution - incubate for 10 min
4. Add Stop solution
5. Read the results on a plate reader at 450 nm
Prekallikrein 1B (KLKB1) is a 88 kDa serine protease involved in blood coagulation, kinin generation, fibrinolysis and inflammation. Prekallikrein 1B is synthesized in the liver and circulates in the blood stream as a free zymogen or in a complex with high molecular weight kininogen. Prekallikrein 1B has a signal peptide (aa1-19) and a pro form (aa20-638). It is composed of a heavy chain and a light chain which are cleaved upon activation.

Biological function summary
Plasma Kallikrein 1B participates in the cleavage of high molecular weight kininogen (HMWK) to release bradykinin a potent vasodilator. This protein is not typically part of a stable macromolecular complex but interacts transiently with other proteins during its activation and function. Its action facilitates increased vascular permeability which plays a role in inflammatory and coagulation responses.
Pathways
Plasma Kallikrein 1B mainly operates within the kallikrein-kinin system and the intrinsic coagulation pathway. It activates Factor XII (Hageman factor) which serves as an upstream regulator in the coagulation cascade. Additionally it connects to other proteins like prekallikrein and high molecular weight kininogen in these pathways coordinating responses in inflammation and blood pressure control.
