Product details

Abcam's proBNP Human ELISA Kit (ab193712) is an in vitro enzyme-linked immunosorbent assay for the quantitative measurement of Human proBNP in serum, plasma and cell culture supernatants.

This assay employs an antibody specific for Human proBNP coated on a 96-well plate. Standards and samples are pipetted into the wells and the immobilized antibody captures proBNP present in the samples. The wells are washed and biotinylated anti-Human proBNP antibody is added. After washing away any unbound biotinylated antibody, an HRP-conjugated streptavidin is pipetted to the wells. After incubation, the wells are again washed, followed by the addition of a TMB substrate solution to the wells. Color will develop in proportion to the amount of proBNP bound in each well. Addition of the Stop Solution will change the color from blue to yellow, and the intensity of the color is measured at 450 nm.

The proBNP also known by names like NT-proBNP and brain natriuretic peptide precursor is a peptide that plays an important role as a cardiac biomarker. This protein has a molecular mass of about 13 kDa and is expressed primarily in cardiac tissues especially the ventricles. ProBNP is synthesized as a precursor and then processed to produce active BNP (B-type natriuretic peptide) and the inactive part NT-proBNP which can be used for diagnosis purposes.
Biological function summary

ProBNP contributes significantly to cardiovascular homeostasis by promoting natriuresis and diuresis which help in reducing blood pressure and volume. It does not form part of a larger protein complex but functions both in the formation of its active peptide BNP and its inactive fragment NT-proBNP. By exerting these effects proBNP influences fluid balance in the body and plays a critical role in maintaining heart function.

Pathways

ProBNP engages in the natriuretic peptide pathways and cyclic GMP signaling. It cleaves to generate BNP which interacts with NPR-A (natriuretic peptide receptor A) on the cell surface leading to increased production of cyclic GMP. This signaling further coordinates with essential proteins like ACE (angiotensin-converting enzyme) and angiotensin II which are fundamental in blood pressure regulation and cardiovascular function.

ProBNP levels correlate strongly with heart failure and acute coronary syndromes. Elevated proBNP or NT-proBNP often indicates stress on the heart due to volume overload or pressure signifying heart malfunction. BNP actively participates in counteracting the renin-angiotensin-aldosterone system (RAAS) helping mitigate the effects of heart failure when its function is not competing with excessive pathological levels of proteins like angiotensin II.
